摘 要:【目的】褐化是植物组织培养中酚类物质被氧化成黑褐色的醌类物质的过程,是影响植物组织培养中愈伤组织诱导率的关键影响因素,该试验是为了研究不同类型的防褐剂对其的抑制效果,并为后续‘中矮1号’的离体快繁研究解决瓶颈问题。【方法】试验采用AgNO_(3)、活性炭(AC)、Na_(2)S_(2)O_(3)、聚乙烯吡络烷酮(PVP)、抗坏血酸(维生素C)5种防褐剂对‘中矮1号’叶片进行防褐化研究。【结果】相比CK组,添加防褐剂可有效缓解组培过程中外植体的褐化情况。其中,维生素C处理组的防褐效果最好,褐化率均在50.00%以内,0.05 g/L的维生素C抗褐化效果最佳,褐化率仅为18.75%;8.0 g/L的PVP效果最差,褐化率高达71.25%,且培养基受浸染褐化严重。【结论】褐化现象的产生有内外多重因素,包括外植体的基因型、母体年龄、取材部位、取材时间及大小等,褐色物质在培养基中的积累也是导致外植体褐变率增加的一个重要因素,MS和1/2MS等固体培养基不利于醌类等毒害物质的扩散,而且会导致其短时间内毒素高浓度聚积,更加剧了外植体的褐化。【Objective】Browning is a process of phenolic substances being oxidized into black-brown quinones,which is a key factor affecting the induction rate of callus in plant tissue culture.This experiment is conducted to study the inhibition effect of different types of anti-browning agents,and to solve the bottleneck problem for the subsequent in vitro rapid propagation research of‘Zhongai 1’.【Methods】AgNO 3,activated carbon(AC),Na_(2)S_(2)O_(3),polyvinylpyrrodanone(PVP)and ascorbic acid(vitamin C)were used to prevent the browning of‘Zhongai 1’leaves.【Results】Compared with CK,browning control agent can effectively alleviate the browning of plants in tissue culture.Among them,VC had the best anti-browning effect,the browning rate was all less than 50.00%,and 0.05g/L VC had the best anti-browning effect with a lowest rate of 18.75%.8.0 g/L PVP was the worst,the Browning rate was highest with a rate of 71.25%,and the medium was seriously stained,too.【Conclusion】The browning phenomenon is caused by multiple internal and external factors,including the genotype of explants,maternal age,sampling site,sampling time and size,etc.The accumulation of brown substance in the medium is also an important factor leading to the increase of the browning rate.Solid medium such as MS and 1/2MS is not conducive to the diffusion of toxic substances such as quinones.Moreover,it will lead to the accumulation of high concentration of endotoxin in a short time,which will aggravate the browning of explants.
