安肠汤对重症溃疡性结肠炎大鼠血浆及粪便中腺嘌呤、乳酸的浓度及肠道梭菌属丰度的影响

摘  要:目的探讨安肠汤通过调控腺嘌呤、乳酸等代谢产物来影响梭菌属等肠道菌群从而治疗重症溃疡性结肠炎(ulcerative colitis,UC)大鼠的作用机制。方法将35只Wistar雄性大鼠随机分为空白组、模型组、美沙拉嗪组、丽珠肠乐组、安肠汤高、中、低剂量组各5只,采用2、4、6—三硝基苯磺酸(TNBS)诱导法构建重症溃疡性结肠炎的大鼠模型。造模后空白组和模型组每只大鼠予以3 mL生理盐水灌胃,每天早晚两次,连续7天,治疗组每只灌胃量是6 mL/(kg·天)(临床等效量的5倍)。复苏后每天早、晚2次,各灌胃药液+生理盐水共3 mL,持续10天。在第10天处死同时各组大鼠,剖腹取腹主动脉血离心取出血浆及病变肠道中的粪便,分别采用高效液相色谱检测各组大鼠血浆中腺嘌呤、乳酸的浓度,及各组大鼠粪便中腺嘌呤、乳酸的浓度,并采用16sDNA测序检测UC大鼠粪便中梭菌属丰度。结果(1)血浆中腺嘌呤、乳酸的浓度:空白组与模型组比较,空白组血浆中腺嘌呤及乳酸浓度显著下调(P<0.05),经美沙拉嗪、丽珠肠乐、安肠汤中剂量组及安肠汤高剂量组介入后,美沙拉嗪组、丽珠肠乐组、安肠汤中剂量组及安肠汤高剂量组与模型组比较,血浆中腺嘌呤及乳酸浓度显著下调(P<0.05)。(2)粪便中腺嘌呤、乳酸的浓度:空白组与模型组相比较,空白组粪便中腺嘌呤及乳酸浓度显著下调(P<0.05)。经美沙拉嗪、丽珠肠乐、安肠汤高剂量组介入后,美沙拉嗪组、丽珠肠乐组及安肠汤高剂量组与模型组比较,粪便中腺嘌呤及乳酸浓度显著下调(P<0.05)。另安肠汤中、高剂量组与安肠汤低剂量组相比较,安肠汤中、高剂量组粪便中腺嘌呤浓度显著下调(P<0.05);安肠汤中剂量组与模型组相比较,安肠汤中剂量组粪便中腺嘌呤浓度显著下调(P<0.05)。(3)粪便中梭菌属浓度:安肠汤中剂量组与空白组相比较,安肠汤中剂量组粪便中梭菌�Objective To investigate the effects of Anchang decoction on the intestinal flora of clostridium species by regulating the metabolism of Adenine and lactic acid in severe ulcerative colitis rats.Methods Thirty-five male Wistar rats were randomly divided into blank group,model group,mesalazine group,Lizhuchangle group,high,middle and low dose of Anchangtang groups with 5 rats in each group,the model of severe ulcerative colitis was induced by 2,4,6-trinitrobenzene sulfonic acid(TNBS).The rats in the blank group and the model group were fed with 3 mL normal saline twice a day for 7 days,in the treatment group,each rat was given 6 mL/(kg/d)(5 times of clinical equivalent)by gavage,twice a day in the morning and evening after resuscitation,and each rat was given 3 mL of medicine plus normal saline for 10 days.On the 10th day,the rats in each group were killed and the plasma and feces in the diseased intestine were extracted by centrifugation from abdominal aorta blood.The concentration of Adenine and lactic acid in the plasma of the rats in each group was detected by High-performance liquid chromatography method,and the concentrations of Adenine and lactic acid in feces of rats in each group,and the abundance of clostridium in feces of UC rats was detected by the method of 16sDNA Results(1)The concentration of adeine and lactic acid in plasma:compared with the model group,the concentration of Adenine and lactic acid in the blank group was significantly decreased(P<0.05),after intervention by Mesalazine,Lizhuchangle,Anchangtang middle dose group and Anchangtang high dose group,mesalazine group,lizhuchangle group,Anchangtang middle dose group and Anchangtang high dose group were compared with the model group,the concentration of ADENINE and lactic acid in plasma was significantly decreased(P<0.05).(2)The concentration of Adenine and lactic acid in feces:compared with the model group,the concentration of Adenine and lactic acid in feces of the blank group was significantly decreased(P<0.05).After intervention with Mesal
