中药调控Rho/ROCK信号通路抑制肿瘤转移的研究进展

摘　　要：Ras同源基因(Ras homolog gene,Rho)/Rho相关螺旋卷曲蛋白激酶(Rho-associated coiled coil-forming protein kinase,ROCK)信号通路与肿瘤的发生发展有密切联系,Rho/ROCK通路被异常激活后Rho/Rac蛋白与下游效应子合作并激活特定的信号网络,可直接或间接影响癌细胞的运动,促进肿瘤的转移进程。近年来,许多研究表明具有清热解毒,活血化瘀的中药复方抗肿瘤疗效显著,并且单味中药的配伍在抗肿瘤中能起到协同作用,延缓肿瘤进展。本文基于Rho/ROCK信号通路介导肿瘤转移的理论视角,将中药复方及单味中药作用于Rho/ROCK通路的具体机制进行总结,发现中医药经Rho/ROCK信号通路对肿瘤转移产生疗效的可能机制包括:抑制肿瘤血管生成拟态、降低血管通透性、减少癌细胞的增殖、促进癌细胞凋亡、恢复细胞极性并诱导细胞骨架改变、减少细胞外基质降解抑制癌细胞侵袭。此外,将本文提及的中药复方和单体总结成表,以方便读者查阅。本文针对目前研究领域的现状进行分析与评价,旨在为研究提供一定的参考。Ras homolog gene(Rho)/Rho-associated coiled coil-forming protein kinase(ROCK)signal pathway is closely related to the occurrence and development of tumors.After Rho/ROCK pathway is abnormally activated,Rho/RAC protein cooperates with downstream effects and activates specific signal networks,which can directly or indirectly affect the movement of cancer cells and promote the metastasis process of tumors.In recent years,many studies have shown that traditional Chinese medicine plays an important role in anti-tumor,which can clear heat and detoxify,promote blood circulation and remove blood stasis,and the combination of single Chinese medicine can play a synergistic role in anti-tumor,delaying tumor progress.Based on the theoretical perspective of Rho/ROCK signal pathway mediated tumor metastasis,this paper summarized the specific mechanisms of Chinese herbal compound and single Chinese herbal medicine acting on Rho/ROCK pathway,and found that the possible mechanisms of Chinese medicine's efficacy on tumor metastasis through Rho/ROCK signal pathway include:inhibition of tumor vasculogenic mimicry,reduction of vascular permeability,reduction of cancer cell proliferation,promotion of cancer cell apoptosis,recovery of cell polarity and induction of cytoskeleton change reduce the degradation of extracellular matrix and inhibit the invasion of cancer cells.In addition,the traditional Chinese medicine compounds and monomers mentioned in this article are summarized into a table for the convenience of readers.This paper analyzes and evaluates the current situation of the research field in order to provide some references for the research.
