摘 要:目的基于BEAS-2B细胞筛选天山花楸抗哮喘活性提取部位,并对其化学成分进行分析。方法采用系统溶剂提取法制备天山花楸不同溶剂提取物;采用脂多糖诱导BEAS-2B细胞构建体外炎症模型,ELISA法检测细胞培养上清液中IL-1β和IL-8水平,筛选天山花楸抗哮喘的活性提取部位;基于UPLC-QE-MS技术对天山花楸抗哮喘活性提取部位进行成分分析。结果ELISA法检测结果表明天山花楸乙酸乙酯、正丁醇和水提取物均可以抑制脂多糖诱导的BEAS-2B细胞分泌IL-1β和IL-8,乙酸乙酯部位抑制效果更为显著;利用UPLC-QE-MS技术对乙酸乙酯部位进行成分分析,通过分析各成分的质荷比及二级碎片离子信息,对其中11个化学成分的结构及其裂解途径进行了推导,包括黄酮类、苯丙素类、酚类及氰苷类化合物。结论本研究对天山花楸乙酸乙酯提取物进行成分分析,为研究天山花楸抗哮喘活性的物质基础提供理论依据。Objective To screen the anti-asthma active extracts from Sorbus tianschanica Rupr.based on BEAS-2B cells and to analyze their chemical compositions.Methods Different solvent extracts from Sorbus tianschanica Rupr.were obtained by systematic solvent extraction method.LPS-induced BEAS-2B cells were used to construct an in vitro inflammation model,and the levels of IL-1β and IL-8 in the cell culture supernatant were detected by ELISA to screen the anti-asthma active extracts of Sorbus tianschanica Rupr..The compositions of the anti-asthma active extracts of Sorbus tianschanica Rupr.were analyzed with UPLC-QE-MS.Results ELISA showed that ethyl acetate extract,n-butanol extract and water extract inhibited the secretion of IL-1β and IL-8 in LPS-induced BEAS-2B cells,and the inhibitory effect of ethyl acetate extract was more significant.The compositions of the ethyl acetate site were analyzed by UPLC-QE-MS,and the structures of the 11 chemical components and their cleavage pathways were deduced from the mass-to-charge ratio and secondary fragment ion information,including flavonoids,phenylpropanoids,phenols and cyanogenic glycosides.Conclusion The composition analysis of the ethyl acetate extract from Sorbus tianschanica Rupr.helps provide a theoretical basis for the material basis for the anti-asthmatic activity of Sorbus tianschanica Rupr..
