摘  要:目的运用网络药理学及生物信息学探讨肾衰泄浊汤治疗慢性肾衰竭的机制。方法通过TCMSP、ETCM、GeneCards、GEO等数据库筛选肾衰泄浊汤的活性成分、靶点及疾病靶点,运用WPS、Cytoscape3.9软件、STRING数据构建药物-成分-靶点-疾病关系网络及PPI网络,将共有基因及差异基因进行GO和KEGG功能富集分析,运用分子对接方法加以验证。结果筛选得到有效成分38种,共同基因112个。GSE37171数据集符合条件,获得113个差异基因。富集分析显示主要涉及通路为IL-17信号通路、脂质与动脉粥样硬化等。结论肾衰泄浊汤以槲皮素等多种成分,通过AKT1、VEGFA等靶点,调节IL-17信号通路、脂质与动脉粥样硬化等通路,发挥对慢性肾衰竭的治疗作用。Objective To explore the mechanism of Shenshuai Xiezhuo Decoction for the treatment of chronic renal failure based on network pharmacology and bioinformatics.Methods The active components,targets,and disease targets of Shenshuai Xiezhuo Decoction were screened from TCMSP,ETCM,GeneCards,GEO,and other databases.WPS,Cytoscape 3.9 software and STRING data were applied to construct drug-components-target-disease network and PPI network.GO and KEGG enrichment analysis were performed for common genes and differential genes,which were confirmed by molecular docking methods.Results Totally 38 active components with 113 common genes were obtained.The GSE37171 dataset was eligible and 112 differential genes were obtained.Enrichment analysis revealed that IL-17 signaling,lipids,and atherosclerosis pathways were mainly involved.Conclusion With quercetin and other components,through AKT1,VEGFA and other targets,Shenshuai Xiezhuo Decoction can regulate IL-17 signal pathway,lipid and atherosclerosis and other pathways to play a therapeutic role in chronic renal failure.
