摘  要:目的探讨丹参通络解毒汤联合甲基转移酶样3(METTL3)对缺氧/复氧(H/R)大鼠心肌微血管内皮细胞(CMECs)血管新生的影响及可能作用机制。方法建立H/R损伤CMECs模型,将细胞分为空白组、模型组、沉默组、过表达组、中药+沉默组、中药+过表达组,细胞划痕实验观察CMECs迁移情况,CCK-8法检测CMECs增殖能力,RT-PCR检测CMECs血管内皮生长因子(VEGF)mRNA表达,Western blot检测内皮型一氧化氮合酶(eNOS)蛋白表达。结果与空白组比较,模型组CMECs迁移率、细胞活力明显降低,VEGF mRNA和eNOS蛋白表达明显降低(P<0.01);与模型组比较,沉默组CMECs迁移率、细胞活力明显降低,VEGF mRNA表达明显降低(P<0.05),过表达组、中药+沉默组、中药+过表达组CMECs迁移率、细胞活力明显增加,VEGF mRNA和eNOS蛋白表达明显升高(P<0.05,P<0.01);与沉默组比较,中药+沉默组CMECs迁移率、细胞活力明显增加,VEGF mRNA和eNOS蛋白表达明显升高(P<0.01);与过表达组比较,中药+过表达组CMECs迁移率、细胞活力明显增加,VEGF mRNA和eNOS蛋白表达明显升高(P<0.05)。结论丹参通络解毒汤联合METTL3可促进H/R损伤CMECs增殖,提高CMECs迁移率,促进血管新生,保护受损CMECs,其机制可能与上调VEGF、eNOS有关。Objective To investigate the effects and possible mechanism of Danshen Tongluo Jiedu Decoction combined with methyltransferase-like 3(METTL3)on angiogenesis of hypoxia/reoxygenation(H/R)myocardial microvascular endothelial cells(CMECs)of rats.Methods H/R injury CMECs model was established,the cells were divided into blank group,model group,silencing group,overexpression group,Danshen Tongluo Jiedu Decoction(TCM)+silencing group,and TCM+overexpression group.Cell scratch test was used to observe the migration of CMECs,and CCK-8 method was used to detect the proliferation of CMECs,RT-PCR was used to detect the expression of vascular endothelial growth factor(VEGF)mRNA in CMECs,and Western blot was used to detect the expression of endothelial nitric oxide synthase(eNOS)protein.Results Compared with the blank group,the migration rate and cell viability of CMECs in the model group were significantly decreased,the expressions of VEGF mRNA and eNOS protein significantly decreased(P<0.01).Compared with the model group,the migration rate and cell viability of CMECs and VEGF mRNA expression in silencing group significantly decreased(P<0.05),the migration rate and cell viability of CMECs in overexpression group,TCM+silencing group and TCM+overexpression group significantly increased,the VEGF mRNA and eNOS protein expression significantly increased(P<0.05,P<0.01).Compared with the overexpression group,the migration rate and cell viability in TCM+overexpression group significantly increased,the expression of VEGF mRNA and eNOS protein increased significantly(P<0.05).Conclusion Danshen Tongluo Jiedu Decoction combined with METTL3 can significantly promote the proliferation of CMECs in H/R rats,improve the mobility of CMECs,promote angiogenesis,and protect damaged CMECs.The mechanism may be related to the up-regulation of VEGF and eNOS.
