摘 要:目的探讨扶正祛邪方调节DGKα-PA信号轴对人肺癌H292细胞增殖、迁移的影响及可能机制。方法将H292细胞分为对照组和扶正祛邪方低、中、高浓度组,扶正祛邪方低、中、高浓度组分别予3、6、12 mg/mL扶正祛邪方干预48 h,对照组予等体积培养基常规培养。克隆形成实验观察H292细胞增殖情况,细胞划痕实验、Transwell实验观察细胞迁移和侵袭情况,ELISA检测细胞磷脂酸(PA)含量,Western blot检测细胞二酰甘油激酶α(DGKα)、蛋白激酶B(AKT)、p-AKT和雷帕霉素机械靶蛋白(mTOR)表达。结果与对照组比较,扶正祛邪方低、中、高浓度组细胞克隆形成数明显减少(P<0.05,P<0.01),细胞迁移率明显降低(P<0.01),侵袭细胞数明显减少(P<0.05,P<0.01),细胞PA含量明显减少(P<0.01),DGKα、p-AKT、mTOR蛋白表达明显降低(P<0.05,P<0.01),且以扶正祛邪方高浓度组抑制作用最明显(P<0.01)。结论扶正祛邪方可能通过DGKα-PA信号轴抑制人肺癌H292细胞增殖、迁移,其机制与抑制下游AKT/mTOR信号通路有关。Objective To investigate the effects and possible mechanism of Fuzheng Quxie Prescription on regulating DGKα-PA signaling axis on the proliferation and migration of human lung cancer H292 cells.Methods H292 cells were divided into control group and Fuzheng Quxie Prescription low-,medium-and high-dosage groups.The Fuzheng Quxie Prescription low-,medium-and high-dosage groups were treated with 3,6 and 12 mg/mL Fuzheng Quxie Prescription for 48 hours,and the control group was routinely cultured with an equal volume of medium.Proliferation of H292 cells were observed by colony formation assay;cell scratch test and Transwell assays were used to observe cell migration and invasion;ELISA was used to detect the content of phosphatidylic acid(PA)in H292 cells;the protein expression of diacylglycerol kinaseα(DGKα),protein kinase B(AKT),p-AKT and mechanistic target of rapamycin(mTOR)were detected by Western blot.Results Compared with the control group,the number of cell clones in the Fuzheng Quxie Prescription low-,medium-and high-dosage groups decreased significantly(P<0.05,P<0.01),the cell migration rate significantly decreased(P<0.01),the number of invasive cells significantly decreased(P<0.05,P<0.01),the content of PA in cells significantly reduced(P<0.01),the protein expressions of DGKα,p-AKT and mTOR significantly decreased(P<0.05,P<0.01).The Fuzheng Quxie Prescription high-dosage group had the most obvious inhibitory effect(P<0.01).Conclusion Fuzheng Quxie Prescription can inhibit the proliferation and migration of human lung cancer H292 cells through the DGKα-PA signaling axis,and its mechanism may be related to the inhibition of downstream AKT/mTOR signaling pathway.
