摘 要:目的观察连草泻痢胶囊对溃疡性结肠炎(UC)大鼠内质网应激及炎症反应的影响,探讨其治疗UC的机制。方法40只雄性SD大鼠随机分为空白组、模型组、连草泻痢胶囊组和美沙拉嗪组,每组10只。除空白组外,其余各组采用葡聚糖硫酸钠溶液自由饮用法构建UC大鼠模型,连草泻痢胶囊组和美沙拉嗪组同时予相应药物干预,连续7 d。测量大鼠结肠长度,对大鼠进行疾病活动指数(DAI)评分,HE染色观察结肠组织病理形态,ELISA检测血清白细胞介素(IL)-1β、IL-17、γ干扰素(IFN-γ)含量,Western blot检测结肠组织葡萄糖调节蛋白78(GRP78)、CHOP、Bax、Bcl-2、半胱氨酸蛋白酶-1(Caspase-1)、核因子(NF)-κB p65、p-NF-κB p65、NF-κB抑制因子(IκB)α、p-IкBα、蛋白激酶R样内质网激酶(PERK)、p-PERK、eIF2α、p-eIF2α、转录激活因子(ATF)4、肌醇需求激酶(IRE)1α、p-IRE1α、xBP1s和Cleaved ATF6蛋白表达。结果与空白组比较,模型组大鼠结肠长度缩短,DAI评分和结肠组织炎症评分升高,血清IL-1β、IL-17、IFN-γ含量增加,结肠组织GRP78、CHOP、Bax、Caspase-1、p-NF-κB p65、p-IкBα、p-PERK、p-eIF2α、ATF4、p-IRE1α、xBP1s和Cleaved ATF6蛋白表达升高,Bcl-2蛋白表达和Bcl-2/Bax比值明显降低,差异均有统计学意义(P<0.01);与模型组比较,连草泻痢胶囊组大鼠结肠长度增加,DAI评分和结肠组织炎症评分降低,血清IL-1β、IL-17、IFN-γ含量减少,结肠组织GRP78、CHOP、Bax、Caspase-1、p-NF-κB p65、p-IкBα、p-PERK、p-eIF2α、ATF4、p-IRE1α、xBP1s和Cleaved ATF6蛋白表达降低,Bcl-2蛋白表达和Bcl-2/Bax比值升高,差异均有统计学意义(P<0.01)。结论连草泻痢胶囊能通过抑制内质网应激改善UC大鼠肠道炎症。Objective To investigate the effects of Liancao Xieli Capsules on endoplasmic reticulum stress and inflammatory response in ulcerative colitis(UC)rats;To exploring its mechanism of treating UC.Methods Totally 40 SD male rats were randomly divided into blank group,model group,Liancao Xieli Capsules group and mesalazine group,with 10 rats in each group.Except for the blank group,the rat model of UC was established in the other groups by free drinking method of dextran sodium sulfate solution,during which the Liancao Xieli Capsules group and the mesalazine group were given corresponding drugs for intervention,for successive 7 d.The length of the colon in rats was measured,and the DAI score was performed,HE staining was used to observe the pathological morphology of colon tissue,ELISA was used to detect the contents of IL-1β,IL-17,IFN-γin serum,Western blot was used to detect the expression of GRP78,CHOP,Bax,Bcl-2,Caspase-1,NF-κB p65,p-NF-κB p65,IκBα,p-IкBα,PERK,p-PERK,eIF2α,p-eIF2α,ATF4,IRE1α,p-IRE1α,xBP1s,and Cleaved ATF6 protein in colon tissue.Results Compared with the blank group,the length of colon was significantly shortened in the model group,the DAI score and colon tissue inflammation score significantly increased,the contents of IL-1β,IL-17 and IFN-γin serum increased,the protein expression of GRP78,CHOP,Bax,Caspase-1,p-NF-κB p65,p-IкBα,p-PERK,p-eIF2α,ATF4,p-IRE1α,xBP1s and Cleaved ATF6 in colon tissue significantly increased,the expression of Bcl-2 protein and the ratio of Bcl-2/Bax significantly decreased,with statistical significance(P<0.01).Compared with the model group,the length of colon increased in the Liancao Xieli Capsules group,the DAI score and colon tissue inflammation score decreased,the contents of serum IL-1β,IL-17,IFN-γdecreased,the expression of GRP78,CHOP,Bax,Caspase-1,p-NF-κB p65,p-IкBα,p-PERK,p-eIF2α,ATF4,p-IRE1α,xBP1s and Cleaved ATF6 protein in colon tissue decreased,the expression of Bcl-2 protein and the ratio of Bcl-2/Bax increased,with statistical signifi
