摘  要:为了解蓝孔雀(Pavo cristatus)肠道菌群中整合子及其基因盒的分布特征,以2个动物园蓝孔雀粪便为材料,采用CTAB法提取肠道菌群宏基因组DNA,共计139份;通过PCR法扩增整合子、基因盒,选取部分PCR产物测序。结果显示:整合子intI1、intI2和intI3检出率分别为86.33%、69.78%和11.51%;不同大小的基因盒条带有18种,共51种组合模式。研究表明蓝孔雀种群中整合子流行性较高,且Ⅰ类整合子携带的基因盒具有高度异质性,反映了蓝孔雀种群潜在多重耐药的复杂性。测序结果显示蓝孔雀携带的耐药基因主要为甲氧苄啶类(dfrA27)和氨基糖苷类(aadA1、aadA2、aac(6′)-Ib和arr),故在用药时应避免这2类药物。In order to understand the distribution characteristics of integrons and their gene cassettes in intestinal flora of Pavo cristatus,139 pieces of metagenomic DNA of intestinal flora were extracted from feces of P.cristatus from two zoos using CTAB method.The integron and gene cassettes were amplified by PCR,and some PCR products were selected for sequencing.The results showed that the detection rates of integron intI1,intI2 and intI3 were 86.33%,69.78%and 11.51%,respectively.There were 18 kinds of strips of varying size in the gene cassettes,with a total of 51 combinations.The integron prevalence was high in the P.cristatus population,and the gene cassettes carried by classⅠintegrons were highly heterogeneous,reflecting the complexity of potential multidrug resistance in the P.cristatus population.Sequencing results showed that drug resistance genes carried by P.cristatus were mainly trimethoprim(dfrA27)and aminoglycosides(aadA1,aadA2,aac(6')-Ib and arr),so that these two drugs should be avoided.
