摘 要:为了明确猪萨佩罗病毒在仔猪腹泻病情中的作用,2016—2020年采集上海市及周边11个猪场腹泻病料,共计1446份。利用RT-PCR方法对其进行PSV筛检,并对阳性样品进行病原分离及病毒特性研究。结果显示:共筛检出256份PSV核酸阳性样品,阳性率17.7%。从阳性样品中成功分离到2株PSV病毒,全基因序列遗传进化分析显示2株PSV分离株聚集在中国PSV毒株集群上,分离株在3D区域4696—5945 bp处存在重组区域。病毒核苷酸分析发现中国PSV毒株与韩国PSV毒株核苷酸相似性为87%—88.5%,与欧洲PSV毒株核苷酸相似性为84.9%—87.3%。对VP1基因氨基酸序列分析发现分离株与参考株存在氨基酸位点突变,中间氨基酸序列保守,无突变及插入和缺少现象。PSV分离株与参考株均含有173位潜在糖基化位点,VP1蛋白分子表面抗原表位分布均匀。该研究可为猪萨佩罗病毒病原学及其疫苗的研制奠定基础。In order to investigate the prevalence of porcine sapelovirus(PSV),a total of 1446 samples of diarrhea materials collected from 11 pig farms in Shanghai and its surrounding areas from 2016 to 2020 were screened for PSV by RT-PCR,and pathogen isolation and whole genome sequence analysis of positive samples were performed to clarify the characteristics of PSV virus.The results showed that 256 samples were positive for PSV.Two PSV strains were successfully isolated from the positive samples.Phylogenetic analysis of genome sequences showed that the two PSV strains clustered in the cluster of Chinese PSV strains,and there was a recombination region in the 3D region of 4696—5945 bp.The nucleotide similarities between PSV strains from China and PSV strains from Korea were 87%—88.5%,and those between PSV strains from China and PSV strains from Europe were 84.9%—87.3%.The amino acid sequence analysis of VP1 gene showed that there were mutations in the amino acid sites between the isolated strain and the reference strain,and the intermediate amino acid sequence was conserved without mutation,insertion and deletion.Compared with the reference strain,PSV isolates strain all contained 173 potential glycosylation sites,and there were uniformly distributed epitopes on the surface of VP1 protein.The results of this study laid the foundation for the etiology and vaccine development of PSV virus in pigs.
