摘 要:为明确喙尾琵琶甲Blaps rynchopetera Fairmaire的蛹不同溶剂提取物的体外抗氧化活,为喙尾琵琶甲全虫态利用价值的开发提供数据支持。采用正己烷、乙酸乙酯、甲醇和水4种不同极性的溶剂提取喙尾琵琶甲蛹,以DPPH法,ABTS法和FRAP法探究不同溶剂提取物体外抗氧化活性强弱。研究结果表明,喙尾琵琶甲蛹的甲醇和水提取物对DPPH自由基和ABTS自由基均有较强清除能力。在提取物浓度为0.5 mg/mL时,水提物和甲醇提取物对两种自由基清除率均达到90%以上,乙酸乙酯提取物和正己烷提取物对两种自由基的清除率较低,清除率均在5%以下,甲醇和水提取物对DPPH自由基清除率的LC50分别为0.144 mg/mL和0.029 mg/mL,对ABTS自由基清除率的LC50分别为和0.0044 mg/mL和0.0082 mg/mL。水粗提物的总抗氧化能力强于甲醇粗提物,浓度为0.5 mg/mL时,甲醇粗提物的总抗氧化能力的FRAP值为0.26,水粗提物的总抗氧化能力的FRAP值0.86。综上所述,研究喙尾琵琶甲蛹水提物的体外抗氧化活性最高,本研究为喙尾琵琶甲蛹体外抗氧化活性评价的首次报道,对喙尾琵琶甲蛹利用价值的研究具有重要意义。In order to determine antioxidant activity in vitro of Blaps rhynchoptera Fairmaire pupa with different polar solvents,and provide data support for utilization value of different stages.Four extracts(n-hexane,ethyl acetate,methanol and distilled water)were used to extract the antioxidant substances from the pupa of Blaps rhynchoptera Fairmaire,DPPH method,ABTS method and FRAP method were sued to explore the antioxidant activity.The research results show that,the extracts of methanol and water showed strong clear effect on DPPH radical and ABTS radical.When the concentration of crude extract was 0.5 mg/mL,the scavenging rate of the radicals was more than 90%by water extract and methanol extract,but below 5%by n-hexane extract and ethyl acetate extract.The LC50 of methanol and water extracts for DPPH radical were 0.144 mg/mL and 0.029 mg/mL,and for ABTS radical were 0.0044 mg/mL and 0.0082 mg/mL.At the same concentration,the FRAP measurement ratio is at 0.26 with methanol extract,and 0.86 with water extract.In conclusion,the water extract has the highest antioxidant activity in vitro.It is the first report on the evaluation of the antioxidant activity of pupa,which was of great significance for studying on pupa of Blaps rhynchoptera Fairmaire.
