摘 要:为了提高抗生素降解菌在堆肥中的降解效率及降低菌株可能引起的抗性基因(ARGs)传播风险,从畜禽粪便好氧堆肥条跺筛选出一株嗜温环丙沙星降解菌株WXX-3,通过模拟堆肥试验探究菌株对环丙沙星的降解效果及ARGs丰度的影响。将菌株以0.05%的接种量加入猪粪,在20~30℃下扩繁8 d,之后加入木屑等调理剂堆肥30 d,环丙沙星的总去除率可达82.9%,比不加菌处理高27.6个百分点。堆肥高温同时抑制了菌体WXX-3的生长,堆肥结束后,未检测出菌体的存在。氟喹诺酮类ARGs及int I1的丰度出现不同程度的消减,其中mex A和qnr A的消减率均达100%,qep A、acr B-01和int I1的消减率分别达17.9%、43.8%和71.7%,明显高于不加菌处理。以上结果表明该菌株可以有效去除环丙沙星,配套的堆肥措施可以发挥该菌株的降解潜力及降低ARGs污染的风险。To enhance the efficiency of antibiotics-degrading bacteria and reduce the risk of the spread of antibiotics resistance genes(ARGs)that may be caused by strains during the composting process,a mesophilic ciprofloxacin-degrading strain WXX-3 was screened from the aerobic composting strips,and a simulated composting experiment was conducted to explore the effects of strain WXX-3 on the ciprofloxacin degradation and ARGs abundance.The strain was added to pig manure at 0.05%of the inoculum,piled up 8 days for its propagation at 20~30℃,and then some conditioner such as wood chips was added to compose for 30 days as conventional compost,the removal rate of ciprofloxacin reached 82.9%,which was 27.6 percentage points higher than the treatment without WXX-3.The high temperature of compost inhibited the growth of WXX-3,after composting,the WXX-3 was not detected.During the composting,the abundance of fluoroquinolone resistance genes and intI1 genes were reduced to different degrees,mexA and qnrA were removed completely,and the contents of qepA,acrB-0 and intI1 were removed 17.9%,43.8%and 71.7%,respectively,which significantly higher than the treatment without WXX-3.The above results indicated that the strain WXX-3 can effectively remove ciprofloxacin,and the supporting composting measures can exert the degradation potential of this strain and reduce the risk of ARGs contamination.
