摘 要:目的:研究健脾宁神颗粒对抽动障碍(TD)小鼠脑内氨基酸类神经递质的影响,探讨健脾宁神颗粒的作用机制。方法:60只雄性ICR小鼠,随机分为正常对照组,模型对照组,健脾宁神颗粒高、中、低剂量组,硫必利阳性对照组。除正常对照组外,其余各组均采用亚氨基二丙腈(IDPN)腹腔注射建立小鼠TD模型。正常对照组和模型对照组给予等体积生理盐水,健脾宁神颗粒高、中、低剂量组分别予45.56、22.78、11.39 g/kg颗粒,硫必利组给药剂量为25 mg/kg,各组小鼠均灌胃给药,每日1次,连续给药4周后,对小鼠进行刻板运动评分,并检测小鼠纹状体内谷氨酸(GLU)、天门冬氨酸(ASP)、γ-氨基丁酸(GABA)含量。结果:与正常对照组比较,模型对照组小鼠脑组织GLU、ASP含量明显升高,GABA含量明显降低,差异具有显著的统计学意义(均P<0.01);与模型对照组比较,健脾宁神颗粒高、中、低剂量组及阳性对照组小鼠脑组织GLU、ASP含量明显降低,GABA含量明显升高,差异具有显著的统计学意义(均P<0.01)。结论:健脾宁神颗粒治疗抽动障碍可能的机制之一是维持兴奋性氨基酸类神经介质和抑制性氨基酸类神经介质的动态平衡。Objective:To explore the mechanism of Jianpi Ningshen Granule by observing the effects of Jianpi Niingshen Granule on amino acid neurotransmitters in the brains of tic disorder mice.Methods:60 Male ICR mice were randomly divided into normal control group,model control group,high,middle,low dose of Jianpi Ningshen Granule groups and Tiapride group with each 10 mice.Except the normal control group,the TD model was established by intraperitoneal injection of Iminodipropanitrile(IDPN)in the other groups.The normal control group and the model control group mice were given the same volume of normal saline,the high,middle and low dose groups of Jianpi Ningshen Granule were given 45.56,22.78,11.39 g/kg Granule,respectively.The Tiapride group was given 25 mg/kg,after 4 weeks of continuous administration,the mice were scored by stereotyped motor score,and the contents of Glu,ASP and Gaba in the striatum were detected.Results:Compared with the normal control group,the contents of Glu and ASP in the brain tissue of the model control group were significantly increased,while the content of Gaba was significantly decreased,with a significant difference between the model control group and the normal control group(P<0.01),the contents of Glu and ASP in the brain tissue of the mice in the high,middle and low dose groups of Jianpi Ningshen Granule and the positive control group were obviously decreased,while the content of Gaba was obviously increased,the differences were significant(P<0.01).Conclusion:One of the possible mechanisms of Jianpi Ningshen Granule in treating tic disorder is to maintain the dynamic equilibrium of excitatory and inhibitory amino acid neurotransmitters.
