摘  要:目的探讨清肺排毒汤对脂多糖(LPS)诱导急性肺损伤(ALI)小鼠的抗炎作用及其机制。方法小鼠随机分为对照组、模型组、地塞米松组及清肺排毒汤低、高剂量组,除对照组外,其余各组采用气管穿刺注射LPS建立ALI模型,造模成功后给予相应药物干预7 d。记录小鼠存活率、肺湿干重比,HE染色观察肺损伤程度,吉姆萨染色法及血液分析仪分类计数肺泡灌洗液(BALF)中炎性细胞,RT-qPCR法检测肺组织TLR4、MyD88、NF-κB mRNA表达,ELISA法检测小鼠血清IL-1β、IL-6、TNF-α水平,Western blot法检测肺组织TLR4、MyD88、p-NF-κB p65、p-IκBα蛋白表达。结果与模型组比较,清肺排毒汤高剂量组小鼠肺水肿和损伤程度,血清、BALF中炎性细胞因子和炎性细胞数量,肺组织TLR4、MyD88、NF-κB mRNA表达和肺组织TLR4、MyD88、p-NF-κB p65,p-IκBα蛋白表达均有不同程度降低(P<0.05,P<0.01)。结论清肺排毒汤可能通过调控TLR4/MyD88/NF-κB信号通路抑制炎症因子和介质释放,减轻LPS诱导的小鼠急性肺损伤。AIM To explore the anti-inflammatory effects and mechanism of Qingfei Paidu Decoction on mice with acute lung injury(ALI)induced by lipopolysaccharide(LPS).METHODS Except for the mice of the control group,the other mice randomly divided into the model group,the dexamethasone group and the low-dose and high-dose groups of Qingfei Paidu Decoction were developed into the ALI models with LPS through trachea puncture followed by the 7 days corresponding drugs administration.Subsequently,the mice had their survival rate and lung wet-dry weight ratio recorded;their extent of lung injury identified by HE staining;their inflammatory cells in BALF counted by Giemsa staining and blood analyzer;their pulmonary mRNA expressions of TLR4,MyD 88 and NF-κB detected by RT-qPCR;their serum levels of IL-1β,IL-6 and TNF-αdetected by ELISA;and their pulmonary protein expressions of TLR4,MyD88,p-NF-κB p65 and p-IκBαdetected by Western blot.RESULTS Compared with the model group,the high-dose Qingfei Paidu Decoction group shared lower degree of pulmonary edema and injury,less number of inflammatory cytokines and cells in serum and BALF,and decreased pulmonary expressions of TLR4,MyD88,NF-κB mRNA and the expressions of TLR4,MyD88,p-NF-κB p65,p-IκBαproteins in varying degrees(P<0.05,P<0.01).CONCLUSION Qingfei Paidu Decoction may alleviate LPS-induced acute lung injury in mice by inhibiting the release of inflammatory factors and mediators via TLR4/MyD88/NF-κB signaling pathway.
