摘  要:目的:探讨翘芩清肺剂调控咳嗽变异性哮喘(CVA)大鼠气道神经源性炎症的作用机制。方法:48只SD大鼠随机分成空白组、模型组、孟鲁司特钠组(1.05 mg/kg)及翘芩清肺剂高、中、低剂量组(26、13、6.5 g/kg),每组8只。采用卵清白蛋白(OVA)联合氢氧化铝[Al(OH)3]致敏并反复多次激发的方法建立大鼠CVA模型,从致敏第2天开始,灌胃给药28 d。各组大鼠HE染色显微镜下观察肺组织的病理变化;双抗夹心ABC-ELISA法测定肺泡灌洗液(BALF)中神经生长因子(NGF)、P物质(SP)的含量变化,免疫组织化学法检测肺组织内NGF、SP的蛋白表达水平。结果:病理结果显示模型组有显著的炎症表现,翘芩清肺剂高、中、低剂量组和孟鲁司特钠组炎症浸润均有不同程度减轻。与空白组比较,模型组大鼠肺组织中NGF、SP的蛋白表达水平明显升高(P<0.01);与模型组比较,翘芩清肺剂高、中、低剂量组和孟鲁司特钠组肺组织中NGF、SP的蛋白表达水平和肺泡灌洗液中的NGF、SP含量均明显降低(P<0.05)。结论:翘芩清肺剂可能通过调节气道神经源性炎症中NGF、SP的蛋白表达水平,从而减轻气道炎症,缓解咳嗽变异性哮喘。Objective:To investigate the mechanism of regulation of airway neurogenic inflammation by Qiaoqin Qingfei agent in rats with cough variant asthma(CVA).Methods:A total of 48 SD rats were randomly divided into blank group,model group,montelukast sodium group(1.05 mg/kg)and high,medium and low dose groups(26,13,6.5 g/kg),with 8 rats in each group.The rat CVA model was established by the method of ovalbumin(OVA)combined with aluminum hydroxide[Al(OH)3)]sensitization and repeated stimulation.From the second day of sensitization,the rat CVA model was given by gavage for 28 days.The pathological changes of lung tissue were observed under microscope by HE staining.The content changes of nerve growth fac‑tor(NGF)and substance P(SP)in alveolar lavage fluid(BALF)were determined by double‑antibody sandwich ABC‑ELISA,and the protein expression levels of NGF and SP in lung tissue were detected by immunohistochemistry.Results:Pathological findings showed significant inflammatory manifestations in the model group,and the inflammatory infiltration in the high‑dose,medium‑dose and low‑dose groups of Qiaoqin Qingfei agent and montelukast sodium groups were alleviated to varying degrees.Compared with blank group,the protein expression levels of NGF and SP in lung tissue of model group were significantly increased(P<0.01).Compared with model group,the protein expression levels of NGF and SP in lung tissue and the contents of NGF and SP in alveolar lavage fluid in high‑dose,medium‑dose and low‑dose groups and montelukast sodium group were significantly decreased(P<0.05).Conclusion:Qiaoqin Qingfei agent may reduce airway inflammation and relieve cough variant asthma by regulating the protein expression levels of NGF and SP in airway neurogenic inflammation.
