摘  要:滤泡辅助性T细胞(Follicular helper CD4 T cells,Tfh)是重要的CD4^(+)T淋巴细胞亚群之一,与多种疾病的发生和发展密切相关,其参与疾病发生的机制尚有待深入研究。该研究将分离小鼠脾脏细胞,应用贝克曼Moflo分选系统分选出CD4^(+)T淋巴细胞。使用细胞因子刺激Tfh细胞分化并检测其分化比例,同时采用流式细胞术检测和分析肉桂活性成分对滤泡辅助性T细胞分化的影响。结果表明,贝克曼Moflo XDP分选系统可以显著提高CD4^(+)T淋巴细胞的纯度,且肉桂活性成分可以显著抑制Tfh细胞分化。为新药在Tfh细胞的开发提供一定实验基础,并且为研发肉桂作为新的增强免疫疗效药物提供可靠的实验依据。Follicular helper CD4 T cells(Tfh)are one of the important CD4+T lymphocyte subsets and are closely related to the occurrence and development of various diseases.The mechanism of their involvement in the occurrence of diseases remains to be further studied.In this study,mouse spleen cells will be isolated and CD4^(+)T lymphocytes will be sorted using the Beckman Moflo XDP sorting system.Cytokines were used to stimulate the differentiation of Tfh cells and the differentiation ratio was detected.Flow cytometry was used to detect and analyze the effect of cinnamon bark extract on the differentiation of follicular helper T cells.The results show that the Beckman Moflo sorting system can significantly improve the purity of CD4^(+)T lymphocytes,and the compounds derived from cinnamon can significantly inhibit the differentiation of Tfh cells,which provides a certain experimental basis for the development of new drugs in Tfh cells,and also provides a reliable experimental basis for the development of cinnamon as a new immune-enhancing drug.
