摘 要:为探究候选基因BMPR-1B、BMP15与东弗里生羊♂和湖羊♀的杂交F_(1)代(东湖F_(1)羊)产羔数间的关联性,评估东弗里生羊作为引入品种的经济利用价值,使用PCR-RFLP技术对东湖F_(1)羊和湖羊的BMPR-1B、BMP15基因多态性与产羔数进行关联分析。结果显示,东湖F_(1)羊与湖羊群体内均未检测出BMP15基因的FecXI突变。BMPR-1B基因FecB突变位点在湖羊群体中检测出AG、GG两种基因型,基因频率分别为0.064和0.936,优势基因型为GG型,等位基因A、G的基因频率分别为0.032和0.968,优势基因为等位基因G;在东湖F_(1)羊群体中检测出AA、AG和GG 3种基因型,基因频率分别为0.146、0.683和0.171,优势基因型为AG型,等位基因A、G的基因频率分别为0.488和0.512。表明对产羔有利的G等位基因可以通过东弗里生和湖羊杂交遗传给后代,可作为其分子育种的辅助选择标记。湖羊AG型与GG型个体产羔数差异不显著(P>0.05),东湖F_(1)羊GG型、AG型个体产羔数极显著高于AA型个体(0.010.05)。结果表明东湖F_(1)羊产羔数与BMPR-1B基因显著相关。In order to explore the correlation between candidate genes BMPR-1B and BMP15 and the litter size of lambs produced in the F1 generation of East Friesland♂and Hu sheep♀(Donghu F1 sheep),and to evaluate the economic utilization value of East Friesland as an introduced breed,PCR-RFLP technique was used to analyze the association between BMPR-1B and BMP15 gene polymorphisms and lamb number in Donghu F1 and Hu sheep.The results showed that FecXI mutation of BMP15 gene was not detected in Donghu F1 and Hu sheep populations.Two genotypes,AG and GG,were detected in the population of Hu sheep with gene frequencies of 0.064 and 0.936,respectively,and the dominant genotype was GG.The gene frequencies of allele A and G were 0.032 and 0.968,respectively,and the dominant gene was G.Three genotypes,AA,AG and GG,were detected in Donghu F1 population,and the gene frequencies were 0.146,0.683 and 0.171,respectively.The dominant genotype was AG,and the gene frequencies of allele A and G were 0.488 and 0.512,respectively.The results indicated that the G allele favorable to lambing could be passed on to offspring through the cross between East Friesland and Hu sheep,and could be used as an auxiliary selection marker in molecular breeding.There was no significant difference in litter size between AG and GG of Hu sheep(P>0.05),but the number of litter size of GG and AG of Donghu F1 was significantly higher than that of AA(0.01<P<0.05),and there was no significant difference between GG and AG of(P>0.05).In conclusion,the results showed that the litter size in Donghu F1 sheep was significantly correlated with the BMPR-1B gene.
