摘 要:2021年9月份湖北省黄冈市某鸡场的产蛋鸡发生以产蛋量下降为主要症状的疾病,为了确定感染的病原,试验采集肝脏、脾脏及大脑等组织进行细菌(鸭源鸡杆菌)、病毒(禽流感病毒、传染性支气管炎病毒)及支原体(鸡毒支原体、滑液囊支原体)的分离培养,采用RT-PCR对分离菌株进行鉴定,并对分离菌株进行生化试验和药敏试验。结果表明:禽流感病毒、传染性支气管炎病毒、鸡毒支原体、滑液囊支原体RT-PCR结果均为阴性;16S rRNA基因和鸭源鸡杆菌的保守基因infB、atpD、rpoB、23S rRNA菌液RT-PCR结果均为阳性;测序结果在NCBI中进行同源性比对,16S rRNA基因与鸭源鸡杆菌的核苷酸相似性大于99%,23S rRNA基因和管家基因ropB、atpD、infB序列与鸭源鸡杆菌的核苷酸相似性均大于99%。该分离菌株能发酵多种糖类,不能发酵乳糖,不能分解尿素和明胶,不产生苯丙氨酸。分离菌株对头孢曲松、阿米卡星、诺氟沙星、氧氟沙星、呋喃妥因、链霉素、复方新诺明及大观霉素高度敏感,而对四环素、克林霉素耐药。说明该鸡场蛋鸡感染了鸭源鸡杆菌,建议该鸡场选择敏感抗生素进行治疗,并且按照药物的休药期合理安排用药。In September 2021,laying hens in a chicken farm in Huanggang City,Hubei Province suffered a disease with a decrease in egg production as the main symptom.In order to determine the pathogen of infection,liver,spleen and brain tissues were collected for isolation and culture of bacteria(Gallibacterium anatis),viruses(avian Influenza A virus,Infectious bronchitis virus)and mycoplasmas(Mycoplasmoides gallisepticum,Mycoplasmopsis synoviae).RT-PCR was used to identify isolated strains,and biochemical test and drug sensitivity test were performed on the isolated strains.The results showed that the RT-PCR results of avian Influenza A virus,Infectious bronchitis virus,Mycoplasmoides gallisepticum and Mycoplasmopsis synoviae were all negative.The bacterial RT-PCR results of 16S rRNA gene and conserved genes infB,atpD,rpoB and 23S rRNA of Gallibacterium anatis were all positive.The sequencing results were compared in NCBI and the nucleotide similarity of 16S rRNA gene of Gallibacterium anatis was greater than 99%,and the nucleotide similarities of 23S rRNA gene and housekeeper genes ropB,atpD and infB sequences were greater than 99%.The isolated strain could ferment a variety of sugars,could not ferment lactose,and could not decompose urea and gelatin,and could not produce phenylalanine.The isolated strain was sensitive to ceftriaxone,amikacin,norfloxacin,ofloxacin,nitrofurantoin,streptomycin,cotrimoxazole,and spectinomycin,but was resistant to tetracycline and clindamycin.The results suggested that the layers of the chicken farm were infected with Gallibacterium anatis,and it was recommended that the farm should choose more sensitive antibiotics for treatment,and reasonably arrange the medication according to the withdrawal period of the drug.
