通城仔猪肝脏和小肠总细胞色素P450的表达规律

摘　　要：通过研究不同月龄、性别的通城仔猪肝脏和小肠微粒体P450活性,为临床安全用药提供理论依据。笔者使用蔗糖密度缓冲液和冷冻差速离心法提取仔猪肝脏和小肠微粒体,采用CO还原差示光谱分析法对微粒体中P450含量进行测定。结果表明,通城猪和杜长大猪3月龄时肝脏和小肠微粒体浓度达到高峰,且杜长大猪肝脏和小肠微粒体浓度均高于通城仔猪(P<0.05)。随着月龄的增长,不同性别的通城猪和三元杂交猪肝脏总细胞色素P450也成比例增长;不同性别的通城猪肝微粒体中P450酶活性均高于三元杂交猪,且雄性通城猪P450含量最高(P<0.05)。This study investigated the differences of microsomal P450 activity in liver and small intestine of Tongcheng piglets with different months of age and sex,so as to provide theoretical basis for clinical safe drug use.Microsomes were extracted from liver and small intestine of piglets using sucrose density buffer and cryodifferential centrifugation,and the content of P450 in microsomes was determined by CO reduction differential spectroscopy.The results showed that the concentrations of microsomes in liver and small intestine of Tongcheng pigs and Du-grown pigs reached the peak at 3 months of age,and the concentrations of microsomes in liver and small intestine of Du-grown pigs were higher than those of Tongcheng piglets(P<0.05).In addition,with the increase of age,the total cytochrome P450 in liver of Tongcheng pigs of diferent genders and terad hybrids also increased in direct proportion.The P450 enzyme activity in liver microsomes of Tongcheng pigs of different genders was higher than that of terad hybrids,and the content of P450 in male Tongcheng pigs was the highest(P<0.05).
