摘 要:目的:使用脓肿分枝杆菌启动子构建抗生素生物传感器。方法:通过使用来自于脓肿分枝杆菌(Mycobacterium abscessus)ATCC19977与头孢西丁耐药的相关基因MAB_2622C和MAB_1858的启动子,以驱动eGFP和lacZ报道基因,构建针对头孢西丁特异性响应的抗生素生物传感器。结果:该抗生素生物传感器可以对1/32最小抑菌浓度有响应,并且随着抗生素浓度越高,lacZ显色越快,吸光度值越低。结论:本实验中的新式传感器具有容易观察、特异性强、灵敏度高等特点,有助于为后续解析分枝杆菌耐药信号通路等分子机制及设计其他新的特异性抗生素传感器提供思路。Objective:To construct an antibiotic biosensor using Mycobacterium abscessus promoter.Methods:An antibiotic biosensor for cefoxitin specific response was constructed by using the promoters of MAB_2622C and MAB_1858 genes from Mycobacterium abscessus ATCC19977 and cefoxitin resistance related genes to drive eGFP and lacZ reporter genes.Results:The antibiotic biosensor could respond to the minimum inhibitory concentration of 1/32,and the higher the antibiotic concentration,the faster the lacZ color develops and the lower the absorbance value.Conclusion:This biosensor has the properties of clear visibility,high specificity,and high sensitivity.This study also provides an understanding of the molecular mechanism of the drug-resistant signal pathways of the Mycobacterium.
