摘 要:为明确MfMel1基因在桃褐腐病菌Monilinia fructicola中的功能,利用生物信息学软件对该基因的结构、所编码蛋白的保守结构域以及进化关系进行分析;通过遗传转化方法对该基因进行敲除,并进行表型分析。结果显示,敲除转化子△MfMel1菌株的菌丝生长速率、孢子萌发率与野生型菌株Bmpc7均无显著差异,但产孢量显著降低。△MfMel1菌株在0.01%SDS、600μg/mL刚果红、150 g/L甘油、200 g/L蔗糖、6 mmol/L H_(2)O_(2)、4%NaCl和1.2 mmol/L山梨醇培养基上的生长速率、菌丝形态均无显著变化,但对200 g/L葡萄糖的渗透胁迫敏感性增强。敲除转化子△MfMel1菌株的α-半乳糖苷酶活性明显降低,对桃果实的致病力显著下降。表明MfMel1基因在桃褐腐病菌中可能通过参与调控α-半乳糖苷酶酶活,从而参与病原菌的致病。In order to illustrate the functions of MfMel1 in pathogen fungus Monilinia fructicola,the gene structure,conserved domain and evolutionary relationships of the predicted protein were analyzed using bioinformatic software.The gene was knocked out through the genetic transformation,and the phenotypes of the mutants were investigated.No significant difference was observed for the mycelial growth and spore germination between knockout transformants△MfMel1 and wild type strain Bmpc7.However,the sporulation of△MfMel1 strains decreased significantly compared to Bmpc7.△MfMel1 strains showed increased sensitivity to 200 g/L glucose,but not to 0.01%SDS,600μg/mL congo red,150g/Lglycerol,200g/Lsucrose,6mmol/LH_(2)O_(2),4%NaCl,or1.2mmol/Lsorbitol.△MfMel1 strains exhibited significantly decreased virulence on peach fruit and reduced activity ofα-galactosidase.The results indicated that MfMel1 was involved in regulating theα-galactosidase activity,thus playing an important role during the infection of M.fructicola on peach fruit.
