摘  要:目的建立一种应用聚合酶链式反应-限制性片段长度多态性技术(PCR-RFLP)鉴别梅花鹿鞭与马鹿鞭的方法。方法根据梅花鹿与马鹿的细胞色素b基因序列的差异,设计并选取DNA限制性内切酶MseI来区分梅花鹿鞭和马鹿鞭。采用改良SDS法提取样本基因组DNA,经PCR扩增后进行RFLP分析,并优化PCR-RFLP反应体系及反应条件,用优化的PCR-RFLP体系对市售鹿鞭样本进行鉴定。结果经PCR-RFLP分析,MseI可切割梅花鹿与马鹿Cyt b片段,前者被切成2个片段,后者被切成3个片段,从而将两者进行鉴别。结论本实验建立的PCR-RFLP方法可准确鉴别梅花鹿鞭与马鹿鞭。Objective To establish a method for identifying sika deer penis and red deer penis by polymerase chain reaction-restriction fragment length polymorphism.Methods The DNA restriction endonuclease MseI was designed and selected to distinguish the sika deer penis from the red deer penis based on the difference in cytochrome b gene sequences between sika deer and red deer.The genomic DNA of samples was extracted by improved SDS method,amplified by PCR and analyzed by RFLP,and the PCR-RFLP reaction system and reaction conditions were optimized.The deer penis samples were identified by the optimized PCR-RFLP system.Results By PCR-RFLP analysis,MseI can cut cytochrome b fragments of sika deer and red deer,the former is cut into two fragments and the latter into three fragments,so as to identify them.Conclusion The PCR-RFLP method established in this experiment can accurately identify the sika deer penis from the red deer penis.
