摘 要:采用正交设计优化胡椒属药用植物SCoT-PCR反应体系,并运用SCoT分子标记对7种胡椒属药用植物31份样品进行遗传多样性研究。结果表明,胡椒属药用植物SCoT-PCR最佳反应体系:总体积20μL,其中10×Buffer 2.0μL,dNTPs 0.20 mmol/L,Mg^(2+) 1.5 mmol/L,引物0.4μmol/L,模板DNA 100 ng,Taq聚合酶2.5 U。扩增总条带数为68条,其中多态性条带为64条,多态性比率为94.12%。31份胡椒属药用植物个体间聚类分析结果显示,31份胡椒属样品划分为2个类群,6个亚群。SCoT可适用于胡椒属药用植物遗传多样性研究。The SCoT-PCR reaction system of medicinal plants of Piper was optimized by orthogonal design,and the genetic diversity of 31 samples of 7 species of medicinal plants of Piper was studied by SCoT molecular markers.The results showed that the best SCoT-PCR reaction system of Piper was as follows:the total volume was 20μL,of which 10×buffer 2.0μL,dNTPs 0.20 mmol/L,Mg^(2+)1.5 mmol/L,primer 0.4μmmol/L,template DNA 100 ng,Taq polymerase 2.5 U.The total number of amplified bands was 68,of which 64 were polymorphic,with a polymorphism rate of 94.12%.The clustering results showed that 31 samples of Piper were divided into 2 groups and 6 subgroups.SCoT can be used to study the genetic diversity of medicinal plants of Piper.
