摘 要:目的基于近红外光谱技术(NIRS)快速检测芪精颗粒中主要药味黄芪和黄精的含量,并比较不同主药含量芪精颗粒治疗白细胞减少症的作用。方法制备不同黄芪和黄精用量的芪精颗粒样本,采用近红外光谱技术结合偏最小二乘法(PLS)建立芪精颗粒中黄芪和黄精用量的近红外快检模型。模型检测采用标准正交变量变换(SNV)预处理,检测波长范围在4000~10000 cm^(-1)。SD大鼠根据白细胞数随机分为:空白对照组、模型组(皮下注射CTX 100 mg/kg)、地榆升白片阳性药对照组、15个不同黄芪和黄精用量给药组,连续给药14 d。在给药后第5,9,11,14天测定血液常规指标、脾腺指数和胸腺指数。结果建立的模型校正均方差(RMSEC)、预测均方差(RMSEP)、交叉验证均方差(RMSECV)、R^(2)值依次为0.0964,0.0846,0.8674,0.9262。给药第14天,与模型组比较,黄芪和黄精用量各6~15 g的给药组血常规指标发生变化,其中黄芪和黄精用量各10~15 g给药组与正常组相比无显著性差异。与模型组比较,各给药组小鼠脾脏和胸腺指数显著性增加(P<0.05),其中黄芪和黄精用量各10~15 g给药组与阳性药组无显著性差异。各指标的总评OD值随黄芪和黄精在处方中的百分含量增高而增高,在百分含量19%处有一个高处的拐点,在百分含量24%处达到最高。结论利用NIRS技术结合多种化学计量学建立的定性定量模型对制剂中黄芪和黄精含量的预测准确度较高。通过近红外模型预测值能间接比较不同主药含量芪精颗粒改善白细胞减少症的作用。Objective To rapidly detect the contents of astragalus and polygonatum in Qijing granules based on near-infrared spectroscopy(NIRS),and compare the effect of different main drug contents of Qijing granules on leukopenia.Methods The samples of Qijing granules with different dosage of astragalus and polygonatum were prepared,and the near-infrared rapid detection model was established to detect the contents of astragalus and polygonatum in Qijing granules using near-infrared spectroscopy and partial least squares(PLS).Model detection was pretreated by standard orthogonal variable transformation(SNV),and the detection wavelength range was set at 4000-10000 cm^(-1).SD rats were randomly divided into 18 groups according to the number of white blood cells:blank control group,model group(subcutaneous injection of 100 mg/kg CTX),positive drug control group(Diyushengbai tablet),15 different dosage groups(Qijing granules with 1-15 g each for astragalus and polygonatum for 14 d).Blood routine indexes,spleen gland index and thymus index were measured at days 5,9,11 and 14 after drug administration.Results The corrected mean square error(RMSEC),predicted mean square error(RMSEP),cross validation mean square error(RMSECV)and R^(2) values of the established model were 0.0964,0.0846,0.8674 and 0.9262,respectively.At day 14,the blood routine indexes in administration groups 6-15(6-15 g each for astragalus and xanthogen)changed compared with model group,and there was no signifcant difference between administration groups(10-15 g each for astragalus and xanthogen)and normal group.The indexes of spleen and thymus of mice were increased in different degrees in each Qijing granule group compared with model group(P<0.05),but there was no signifcant difference between administration groups 10-15(10-15 g each for astragalus and xanthogen)and positive drug group.The overall evaluation OD value of each index increased with the increase of the percentage content of astragalus and polygonatum in the prescription,with a high inflection point a
