中药复方体外药理研究方法思考

摘　　要：中药复方的体外药理机制研究是明确中药复方效应靶点和清晰阐释中医药内涵的关键,然而中药复方不同于单一成分或化学药物,其物质基础不明、效应成分不清和复杂的成分组成等特点使中药复方的体外药理研究方法争议颇多。就细胞实验而言,直接干预方法多采用水煎液或醇提物直接加入细胞培养体系,间接干预方法主要为制备含药血清、含药肠吸收液、含药肝孵育液或含药脑脊液等加入细胞培养体系,亦或是筛选有效活性成分直接进行药理机制研究。然而,每种方法均存在一定的缺点,不能完全清晰地实现中药复方的体外机制研究,因此,针对不同研究需求,选择合适的中药复方体外药理研究方法,通过研究设计规避其缺点,优化出具备较高科学性和可重复性的研究方案是中药复方体外药理研究亟待解决的关键技术问题。The study of the in vitro pharmacological mechanism of Chinese herbal compound plays the key role in clarifying the effect targets of Chinese herbal compound and clearly explaining the connotation of traditional Chinese medicine.However,unlike single component or chemical drugs,Chinese herbal compound is characterized by unknown material basis,unclear effective components and complex component composition,which makes the in vitro pharmacological research methods of Chinese herbal compound quite controversial.In terms of cell experiments,most of the direct intervention methods are water decoction or alcohol extract directly added to the cell culture system,while the indirect intervention methods mainly include drug-containing serum,drug-containing intestinal absorption solution,drug-containing liver incubation solution or drug-containing cerebrospinal fluid,as well as,screening the active ingredients and exploring their pharmacological mechanism directly.However,each method has certain shortcomings,which cannot completely and clearly realize the in vitro mechanism research of Chinese herbal compound.Therefore,according to different research needs,selecting appropriate in vitro pharmacological research methods,avoiding their shortcomings through research design,and optimizing the research scheme with high scientificity and repeatability are the key technical problems that need to be solved urgently in the in vitro pharmacological research of Chinese herbal compound.
