摘  要:目的:探讨甲炎康泰对自身免疫性甲状腺炎(AIT)大鼠保护作用的机制。方法:制作AIT大鼠模型,随机分为模型组、甲炎康泰组,每组10只。另选取10只为正常组。灌胃干预8周后,ELISA检测大鼠血清中甲状腺过氧化物酶抗体(TPOAb)浓度,甲状腺组织进行HE染色、实时荧光定量PCR检测和免疫组化检测。结果:与模型组比较,甲炎康泰组TPOAb浓度显著降低(P<0.05),淋巴细胞浸润减轻;甲炎康泰组大鼠甲状腺组织中维甲酸相关核孤儿受体γt(RORγt)mRNA及蛋白表达显著降低(P<0.01,P<0.05),叉头翼状螺旋转录因子P3(FOXP3)、转化生长因子-β(TGF-β)、Smad3 mRNA及蛋白表达均显著升高(P<0.01,P<0.05)。结论:甲炎康泰可能通过增强甲状腺组织中TGF-β、FOXP3以及Smad3的表达和抑制RORγt的表达以缓解AIT的发生发展。Objective:To investigate molecular mechanism of Jia-Yan-Kang-Tai in autoimmune thyroiditis(AIT).Methods:AIT rat models were established and randomly divided into the model group and the Jia-Yan-Kang-Tai group,10 rats in each group.Ten rats were selected as normal group.Rats in each group were administrated corresponding drugs for 8 weeks.Serum levels of thyroid peroxidase antibody(TPOAb)were measured by ELISA,and thyroid tissues were examined by HE staining,real-time quantitative PCR and immunohistochemistry.Results:The levels of TPOAb(P<0.05)and lymphocyte infiltration in the Jia-Yan-Kang-Tai group were significantly lower than those in the model group.The mRNA and protein expression levels of ROR t in thyroid tissue of the rats in the Jia-Yan-Kang-Tai group were significantly lower than those in the model group(P<0.01,P<0.05).The mRNA and protein expression levels of FOXP3,TGF-β,Smad3 were increased than those in the model group(P<0.01,P<0.05).Conclusion:Jia-Yan-Kang-Tai may alleviate the development of AIT by enhancing the expression of TGF-β,FOXP3 and Smad3 in thyroid tissue and inhibiting the expression of ROR t.
