摘 要:目的:研究右归饮对大鼠激素性股骨头坏死(SANFH)的影响,探讨其作用机制。方法:取30只SD大鼠按随机数字表法分成正常组、模型组及右归饮组,每组10只。除正常组外,其余各组大鼠采用脂多糖(LPS)联合激素法制备SANFH模型。右归饮组在造模同时灌胃相应剂量的右归饮,1次/天,连续8周,模型组和正常组给予等容积生理盐水。末次给药后,取材并进行血液流变学检测;检测血清总胆固醇(TC)、甘油三酯(TG)、高密度脂蛋白(HDL)、低密度脂蛋白(LDL)水平;采用HE染色观察大鼠股骨头病理学改变;免疫组织化学染色检测观察大鼠股骨头Caspase-3、β-catenin、Wnt3α蛋白表达情况。结果:与模型组比较,右归饮组的全血黏度(低切、中切、高切)及血浆黏度均明显降低(P<0.05或P<0.01);右归饮组血清SOD及GSH-PX含量显著升高(P<0.05),而MDA水平显著降低(P<0.05);股骨头结构及其病理损伤均有一定程度的改善;免疫组织化学染色示,右归饮组Caspase-3阳性细胞百分比显著降低(P<0.01),右归饮组β-catenin、Wnt3α细胞百分比显著升高(P<0.01)。结论:右归饮可通过降低血脂起到预防和缓解股骨头坏死进展的作用,而其作用机制可能与其能激活Wnt/β-catenin通路有关。Objective:To research the effect of Yougui yin on steroid-induced avascular necrosis of femoral head(SANFH)in rats,and to approach its mechanism.Methods:30 SD rats were divided into normal group,model group and Yougui yin group by method of random number table,10 rats in each group.Except normal group,the rats in other groups were made SANFH model by lipopolysaccharide(LPS)and hormone method.Each dosage group of Yougui yin was administered the corresponding dosage of Yougui yin by gavage at the same time of modeling,once a day,continuously for 8 weeks.The model group and normal group were administered the same volume of normal saline.After the last administration,the samples were taken,and the hemorheology was detected.Total cholesterol(TC),trigly ceride(TG),high density lipoprotein(HDL)and low density lipoprotein(LDL)in serum were detected.The pathological changes of femoral head in rats was observed by HE staining.The expressions of Caspase-3,β-catenin and Wnt3αprotein in femoral head of rats were detected and observed by immunohistochemical staining.Results:Compared with the model group:the whole blood viscosity(low-,middle-and high-shear)and plasma viscosity of Yougui yin group were significantly reduced(P<0.05 or P<0.01);the contents of serum SOD and GSH-PX in Yougui yin group were significantly increased(P<0.05),while MDA level was significantly reduced(P<0.05);the structure of femoral head and its pathological damage were improved to some extent;the immunohistochemical staining showed that the progression of femoral head necrosis by reducing blood lipid,and its mechanism may be related to activating signaling pathway of Wnt/β-catenin.
