摘 要:为筛选出对核桃长足象具有高致病性的虫生真菌,从核桃林采集核桃长足象僵虫,在马铃薯葡萄糖琼脂(PDA)培养基上分离纯化得到菌株QZCZX-1,通过光学显微镜和扫描电镜进行形态学观察鉴定,并进行DNA提取、PCR、测序、BLAST比对等分子生物学鉴定,确定其为球孢白僵菌(Beauveria bassiana),同时测定该菌株对核桃长足象的致病性。结果显示:随着孢子悬浮液的菌浓度升高,核桃长足象的校正死亡率增加,半致死时间(LT 50)缩短;菌浓度为108个/mL时,核桃长足象的校正死亡率高达92.59%。由此表明,菌株QZCZX-1对核桃长足象具有较强的致病性。为后续核桃长足象的生物防治提供菌源。In order to screen out the entomogenous fungi with high pathogenicity to Alcidodes juglans Chao,the entomogenous fungi QZCZX-1 was isolated and purified on potato dextrose agar(PDA),the strain was identified as Beauveria bassiana by morphological observation,DNA extraction,PCR,sequencing and BLAST analysis.At the same time,the pathogenicity of the strain to A.juglans was tested.The results showed that the corrected mortality of A.juglans increased and the LT 50 value decreased with the increase of the concentration of spore suspension,when the concentration of the strain was 108ind./mL,the adjusted mortality rate of Alcidodes juglans Chao was 92.59%.The results showed that strain QZCZX-1 had strong pathogenicity to Alcidodes juglans.The fungus source would be provided for biological control of Alcidodes juglans Chao in future.
