摘 要:目的:研究二氧化钛纳米颗粒(TiO2NPs)对小鼠胚胎发育的作用,并探讨其机制。方法:将小鼠随机分为4组,分别为正常对照组、低剂量组、中剂量组、高剂量组,自妊娠第1天(GD1)起至GD17.5,分别灌胃给予TiO2NPs 0、10、50、250(mg/kg/d),并每天测量体重。GD17.5时处死母鼠,立即解剖取心、肝、脾、肺、肾、脑等器官进行称重,剖开子宫取胎鼠拍照,收集组织样本进行蛋白免疫印迹。结果:在GD17.5时,各组妊娠小鼠的体重无显著统计学差异,与正常对照组相比,高剂量组母鼠的肝的器官重量系数明显降低(P<0.05)。随着TiO2NPs剂量增加,可见异常发育胎鼠增多,且高剂量组(P<0.05)胎鼠中NF⁃κB p65蛋白表达水平明显升高。结论:TiO2NPs对小鼠母胎产生不良影响,并能使胎鼠NF⁃κB p65蛋白表达水平升高。Objective:To study the effect of titanium dioxide nanoparticles(TiO2NPs)on mouse embryonic devel⁃opment and to investigate the mechanism.Methods:The mice were randomly divided into four groups:normal control group,low⁃dose group,middle dose group and high⁃dose group.From the first day of pregnancy(GD1)to GD17.5,they were given TiO_(2)NPs 0,10,50 and 250(mg/kg/d)by gavage,and their body weights were measured every day.At GD17.5,the dams were killed and anatomized immediately,and the heart,liver,spleen,lung,kidney,brain,uterus and other organs were taken out for weighing and photographing.Then,the uterus was dissected to take fetal mice for photographing,and tissue samples were collected for western blot.Results:At GD17.5,there was no significant differ⁃ence in the body weight of the dams in each group.Compared with the control group,the organ weight coefficient of the liver of the dams in the high⁃dose group decreased significantly(P<0.05).With the increase of the dosage of TiO_(2) NPs,the number of abnormally developed fetuses increased,and the expression level of NF⁃κB p65 protein in the fetuses of the high dose group(P<0.05)increased significantly.Conclusion:TiO_(2)NPs adversely affected the mothers and fetuses of mice and increased the level of NF⁃κB p65 protein expression in fetuses of mice.
