摘 要:为了明确野生秦岭羚牛(Budorcas taxicolor bedfordi)食道口线虫(Oesophagostomum)的种类与遗传进化关系,对秦岭山区的1只野外死亡羚牛结肠内的食道口线虫利用蠕虫完全剖检法进行采集,初步观察虫体基本形态,并基于ITS rRNA、cox1和nad1基因位点对虫体进行分子生物学鉴定和遗传进化分析。结果发现,共分离得到39条食道口线虫虫体,形态学鉴定符合食道口线虫的典型结构特征;代表性虫体样本的ITS rRNA基因、cox1和nad1部分基因的PCR扩增产物长度分别为913、441、721 bp,其序列与山羊源粗纹食道口线虫(O.asperum,GenBank登录号:KM200806、KM200767、KM200817)相似性分别为99.49%~99.75%、99.43%~100%和98.94%~99.39%,在遗传进化树中位于同一支,而与其他食道口线虫以及蛊口科其他线虫遗传距离较远。结果表明,该只野生秦岭羚牛结肠内的食道口线虫为粗纹食道口线虫。研究结果为秦岭羚牛食道口线虫病的流行病学调查和防控提供了基础数据。The present study aimed to identify and genetically analyze Oesophagostomum sp.from Budorcas taxicolor bedfordi.Oesophagostomum samples in the colon of a dead B.taxicolor bedfordi in Qinling mountainous areas were isolated using complete necropsy.The basic morphology of all samples was preliminary observed.Molecular biological identification and genetic analysis of isolated samples were performed based on the ITS rRNA gene,cox1 partial gene and nad1 partial gene.The results showed that 39 parasites were obtained from the colon.The morphological features of these parasites were consistent with the typical structural characteristics of Oesophagostomum.The PCR amplification products of ITS rRNA gene,cox 1 partial gene and nad 1 partial gene of representative samples were 913 bp,441 bp and 721 bp,respectively.Sequence analysis revealed highest similarities of sequences from obtained parasites for O.asperum(GenBank Numbers:KM200806,KM200767,KM200817),with 99.49%-99.75%,99.43%-100%and 98.94%-99.39%for ITS rRNA gene,cox 1 partial gene and nad 1 partial gene,respectively.Phylogenetic trees showed that representative samples were located in the same branch with O.asperum,but were genetically distant from other worms within Oesophagostomum and other Cyathostomidae.The results indicated that the nematodes isolated from B.taxicolor bedfordi are O.asperum.The present study provides basic data for epidemiological investigation,prevention and control of oesophagostomiasis in B.taxicolor bedfordi.
