摘 要:目的探究红花多糖对盐酸林可霉素引起的小鼠肠道菌群失调的改善效果。方法选取50只昆明小鼠适应性饲养1周后,随机取10只为对照组,其余40只给予盐酸林可霉素连续灌胃3 d,构建肠道菌群失调模型小鼠,然后随机分为模型组、红花多糖组(150 mg/kg)、丽珠肠乐组(7 mg/kg)、自然恢复组,每组各10只。除模型组外,其他各组第4天起灌胃给药,对照组、自然恢复组灌胃等体积的生理盐水,连续灌胃7 d。灌胃结束后,用酶联免疫吸附法(ELISA)对各组小鼠血清白细胞介素6(Interleukin-6,IL-6)和肿瘤坏死因子-α(tumor necrosis factor,TNF-α)进行检测。无菌收集小鼠回盲部内容物对肠道菌群计数。结果小鼠给予一定剂量的盐酸林可霉素后肠杆菌和肠球菌数量显著增加(P=0.000),双歧杆菌和乳酸杆菌数量明显下降(P=0.000,P=0.003);血清中IL-6和TNF-α水平显著提高(P=0.000)。红花多糖干预后,双歧杆菌和乳酸杆菌数量显著增加(P=0.001),肠杆菌和肠球菌数量显著减少(P=0.037,P=0.002);IL-6和TNF-α水平显著降低(P=0.000)。结论红花多糖对小鼠的肠道菌群失调具有调节作用,并使其恢复正常状态。Objective To explore the effect of Safflower Polysaccharides on dysbiosis of murine gut microbiota induced by lincomycin hydrochloride.Methods 50 Kunming mice were adapted feeding for one week.10 of which were randomly selected as the control group and the remaining 40 mice were given lincomycin hydrochloride for 3 days to construct the intestinal flora dysbiosis model mice and then they were randomly divided into model group,safflower polysaccharide group(150 mg/kg),Livzon Changle group(7 mg/kg),and natural recovery group(normal saline),with 10 mice in each group.From the 4th day,except for the model group,the rest of the groups were given drugs or equal volume of normal saline accordingly for 7 days.After the gavage,the serum IL-6 and TNF-αof mice in each group were detected by enzyme-linked immunosorbent assay(ELISA).The mice ileoceca contents were collected to count the intestinal flora.Results After experimental mice were given a certain dose of lincomycin hydrochloride,the numbers of enterobacteria and enterococci significantly were increased(P<0.01),and the contents of bifidobacteria and lactobacilli were decreased(P<0.05,P<0.01).The levels of IL-6 and TNF-αin serum were significantly increased(P<0.01).After the intervention of safflower polysaccharide,the numbers of bifidobacteria and lactobacilli were increased(P<0.01),the numbers of Enterobacter and Enterococcus were decreased(P<0.05,P<0.01),the levels of IL-6 and TNF-αwere significantly decreased(P<0.01).Conclusion Safflower polysaccharide could regulate the intestinal dysbacteriosis in mice to a normal conditions.
