深低温冻存肾肿瘤患者全血细胞RNA提取质量的影响因素

摘　　要：为了研究深低温冻存的全血细胞RNA提取质量的影响因素,本文选择海军军医大学附属长海医院生物样本库收集、冻存的186份肾肿瘤患者的全血细胞标本,采用商用试剂盒提取RNA并测定其纯度。根据不同冻存时间和冻存位置比较各组全血细胞RNA提取质量。结果表明:本组全血细胞RNA的冻存时间中位数为101(62,127.25)d,260 nm和280 nm波长处的吸光度比值(A260/A280)中位数为1.87(1.76,1.92),A260/A230比值中位数为1.72(1.23,1.96),样本RNA提取质量合格率61.3%;冻存时间短的样本RNA提取质量合格率高于冻存时间长的样本,且随着冻存时间变长,A260/A280比值下降。冻存于深低温冰箱上层的样本RNA提取质量合格率高于下层的样本。冻存时间和冻存位置均会对深低温冻存的全血细胞RNA提取质量有影响,应在样本冻存时予以注意。To explore the factors affecting the quality of RNA extraction of cryopreserved whole blood cells from patients with kidney neoplasms,a total of 186 whole blood cells samples of patients with kidney neoplasms,frozen and preserved at clinical biobank of Changhai Hospital,affiliated to the Naval Medical University,are used for quality verification.The quality of RNA extraction in each group is compared.The median freezing-time is 101(62,127.25)days,the median A260/A280 ratio is 1.87(1.76,1.92),and the median A260/A230 ratio is 1.72(1.23,1.96).The total qualified rate of RNA extraction in this group is 61.3%.The qualified rate of RNA extraction in shortterm frozen-preserved group is higher than that in long-term frozen-preserved group.As the freezing-time becomes longer,the A260/A280 ratio decreased.In addition,the qualified rate of RNA extraction of the samples frozen in upper layer is significantly higher when compared with those in lower layer.Both the freezing-time and the storage location influence the quality of RNA extraction of cryopreserved whole blood cells,which should be taken into consideration for sample storage.
