复方熊胆茵陈颗粒对代谢相关脂肪性肝病大鼠HPA轴相关激素影响的研究

摘　　要：目的 研究复方熊胆茵陈颗粒(ACF)对代谢相关脂肪性肝病(MAFLD)大鼠HPA轴相关激素的影响。方法 将51只SD大鼠随机分为模型组11只,正常组、低剂量组、中剂量和高剂量组各10只,正常组给予基础饲料、模型组与各剂量组给予高脂饲料喂养6周。造模成功后,低、中、高剂量组按体质量1 mL/100 g分别予4.73、9.46、18.92 g/L复方熊胆茵陈颗粒药液灌胃,正常组和模型组给予2 mL生理盐水灌胃,共4周。取肝脏行HE染色观察5组大鼠肝组织病理变化,取血清检测5组大鼠血清中丙氨酸氨基转移酶(ALT)、天冬氨酸氨基转移酶(AST)、促肾上腺皮质激素释放激素(CRH)、促肾上腺皮质激素(ACTH)、皮质醇(CORT)水平,qPCR检测5组大鼠肝组织中CRH、ACTH、CORT mRNA相对表达水平,免疫组化检测5组大鼠肝组织中CRH、ACTH、CORT蛋白相对表达水平。结果 (1)与正常组比较,模型组大鼠肝索排列紊乱,脂肪空泡满视野,累及肝小叶大于50%,符合脂肪性肝病病理+++级;与模型组比较,各剂量组大鼠肝细胞的脂肪空泡减少,肝索排列整齐,肝小叶清晰可见。(2)与正常组比较,模型组血清ALT、AST、CRH、ACTH、CORT均显著升高(P均<0.01);与模型组比较,各剂量组大鼠血清ALT、AST、ACTH、CORT均显著降低(P均<0.01),中、高剂量组大鼠血清CRH均显著降低(P<0.05或P<0.01)。(3)与正常组比较,模型组大鼠肝脏CRH、ACTH、CORT mRNA相对表达水平均显著升高(P均<0.01);与模型组比较,低、中、高剂量组大鼠肝脏CRH、ACTH、CORT mRNA相对表达水平均显著降低(P<0.05或0.01)。(4)与正常组比较,模型组大鼠肝脏CRH、ACTH、CORT蛋白相对表达水平均显著升高(P均<0.05);与模型组比较,各剂量组大鼠肝脏CRH、ACTH、CORT蛋白相对表达水平均显著降低(P均<0.05)。结论ACF能够改善MAFLD大鼠肝组织损伤,减少MAFLD大鼠血清及肝组织中CRH、ACTH、CORT的分泌,提示ACF治疗MAFLD的作用机制可能与改�Objective:To study the effect of Compound Xiongdan Yinchen Granules on HPA axis related hormones in rats with metabolic-associated fatty liver disease(MAFLD).Methods:A tatol of 51 SD rats were randomly divided into normal group(n=10),model group(n=11),low-dose group(n=10),medium-dose group(n=10)and high-dose group(n=10).The normal group was fed with basic diet,and the model group and each dose group were fed with high-fat diet for 6 weeks.After successful modeling,the low,medium and high dose groups were given 4.73,9.46 and 18.92 g/L compound Xiongdan Yinchen Granules by intragastric administration according to the body weight of 1 mL/100 g,while the normal and model groups were given 2 mL normal saline by intragastric administration for a total of 4 weeks.HE staining was used to observe the pathological changes of liver tissue of rats.Serum levels of alanine aminotransferase(ALT),aspartate aminotransferase(AST),corticotropin releasing hormone(CRH),adrenocorticotropin(ACTH)and cortisol(CORT)were detected.qPCR and immunohistochemistry were used to detect the expression of CRH,ACTH,CORT mRNA and protein in the liver tissues of rats.Results:①Compared with the normal group,the hepatic cord arrangement of rats in the model group was disordered,the adipose vacuoles filled the visual field,and the involvement of the hepatic lobule was more than 50%,which was consistent with the pathological grade+++of fatty liver disease.Compared with the model group,the adipose vacuoles of hepatocytes in each dose group were reduced,the hepatic cords were arranged neatly,and the hepatic lobules were clearly visible.②Compared with the normal group,the serum levels of AST,ALT,CRH,ACTH and CORT in the model group significantly increased(P<0.01).Compared with the model group,the serum levels of AST,ALT,ACTH and CORT in all dose groups significantly decreased(P<0.01),and the serum levels of CRH in the medium and high dose groups significantly decreased(P<0.05,P<0.01).③Compared with the normal group,the mRNA expression levels of CRH,AC
