维生素D3的摄入对大鼠乳腺增生状态的缓解及干预作用

摘  要:目的建造大鼠乳腺增生模型并使用维生素D3(VD3)进行干预,探讨VD3对乳腺增生的缓解及干预作用。方法SD大鼠35只随机分为对照组、增生组、VD3低、中、高剂量组5组,除对照组外,其余各组建造乳腺增生模型,VD3各组分别同时注射5、10、20μg/kg VD3共30天;实验结束后,测量大鼠乳头直径、高度及乳腺厚度;对乳腺组织进行HE染色,并检测各组乳腺组织中ERα、PR、VDR表达水平。结果经VD3干预后,低、中、高剂量组乳腺厚度分别降低至1.76 mm、1.71 mm、1.86 mm;乳头直径分别降低至1.26 mm、1.27 mm、1.32 mm;乳头高度分别降低至1.46 mm、1.37 mm、1.57 mm,均明显小于增生组的2.24 mm、1.90 mm、2.47 mm,差异均有统计学意义;HE染色显示经VD3干预后乳腺增生状态明显缓解;Western blot显示VD3各组中ERα及PR的表达较增生组均有下降,其差异具有统计学意义(P<0.01);VDR的表达在VD3各组内较对照组均有升高,差异具有统计学意义(P<0.01)。结论经VD3干预后,大鼠乳腺增生状态明显缓解,乳腺组织内ERα、PR表达下降,VDR表达增加。Objective To explore the intervention effect of vitamin D3 on mammary gland hyperplasia.Methods A total of 35 female rats were divided into 5 groups randomly:blank control group,hyperplasia model group,and vitamin D3(VD3)groups of low-dose(LVD,5μg/kg),medium-dose(MVD,10μg/kg),and high-dose(HVD,20μg/kg).Hyperplasia of mammary glands(HMG)model was established in all groups except for the blank control group,then different dose of VD3 was intraperitoneal injected for VD3 groups.After the experiment,the heights and diameters of nipples,and the thickness of the mammary gland of rats were measured.The tissues of mammary glands were analyzed by HE stains,and the total protein expression of ERα,PR,and VDR were measured by western blot.Results After VD3 intervention,the breast thickness decreased to 1.76 mm,1.71 mm and 1.86 mm respectively,and the nipple diameters decreased to 1.26 mm,1.27 mmand 1.32 mm respectively,and the nipple heights decreased to 1.46 mm,1.37 mm,and 1.57 mm respectively,which were significantly lower than those in hyperplasia group(2.24 mm,1.90 mm,2.47 mm).HE staining showed that the mammary gland tissue of rats was significantly relieved after intervention with VD3.Western Blot showed that the expression of ERαand PR in each VD3 group was significantly lower than that in the hyperplasia group,and the expression of VDR in each VD3 group was significantly higher than that in the control group(P<0.01).Conclusion Supplementation of VD3 could markedly decrease the mammary gland thickness,nipple diameter and nipple height of rats.Intervention with VD3 can lead to decreased expression of ERαand PR,in conjunction with the increase of VDR.
