摘 要:为比较实验室分离保存的猪轮状病毒田间株(GZAS2020株)与猪三联腹泻活疫苗NX株的差异,参照已发表的A群猪轮状病毒VP4、VP6、VP7序列设计合成3对扩增全基因的通用引物,分别克隆测序GZAS2020株和NX株的VP4、VP6、VP7全基因。采用DNAStar对GZAS2020株和NX株VP4、VP6、VP7这3个主要基因进行核苷酸、氨基酸同源性分析以及遗传进化树构建,结果显示,GZAS2020株与NX株VP4、VP6、VP7基因核苷酸同源性分别为75.2%、90.0%和79.4%,氨基酸同源性分别为78.5%、74.1%和82.4%,根据G/P分类命名法,GZAS2020株属于G5P[13]。与NX疫苗毒株相比,GZAS2020株的VP4基因在359、360、750 bp处存在C、A、C碱基的缺失,在347、348、570、571、572、579、580、581、757 bp处分别插入了C、C、C、A、A、G、G、G、C碱基;VP6基因不存在插入和缺失;VP7基因在258、1 029 bp处分别存在T、A碱基的插入,266 bp处存在1个C碱基的缺失。结果表明,GZAS2020株和NX株在核苷酸位点上有一定的插入、缺失以及突变,导致编码氨基酸有意义突变,致使蛋白抗原表位出现差异。研究结果可为临床疫苗的选用、诊断方法的建立及猪轮状病毒疫苗候选株的筛选提供参考,也丰富了贵州省猪轮状病毒病的流行病学资料。In order to compare the differences between the porcine rotavirus field strain(GZAS2020 strain)isolated and preserved in our laboratory and the NX strain of the live porcine triple diarrhea vaccine,this study designed and synthesized 3 pairs of universal primers used to amplify the whole genomes,the whole genomes of VP4,VP6 and VP7 of GZAS2020 strain and NX strain were cloned and sequenced respectively.DNAStar was used to analyze the nucleotide and amino acid homologies of the three main genes of VP4,VP6 and VP7 in GZAS2020 and NX strains,and the genetic evolutionary tree was constructed.The results showed that the nucleotide homologies of the VP4,VP6 and VP7 genes of the GZAS2020 strain and the NX strain were 75.2%,90.0%and 79.4%,respectively,and the amino acid homologies were 78.5%,74.1%and 82.4%,respectively.According to the G/P classification nomenclature,the GZAS2020 strain belongs to G5P[13].Compared with the NX vaccine strain,the VP4 gene of the GZAS2020 strain has deletions of C,A and C bases at 359,360,and 750 bp;C,C,C,A,A,G,G,G,and C bases were inserted at 347,348,570,571,572,579,580,581,757 bp;there were no insertions and deletions in the VP6 gene;T and A bases were inserted at 258 and 1029 bp in the VP7 gene,respectively,there was a C-base deletion at 266 bp.The above results indicated that the GZAS2020 strain and the NX strain had certain insertions,deletions and mutations at nucleotide sites,resulting in meaningful mutations in the encoded amino acids,resulting in differences in protein epitopes.The research results can provide reference for the selection of clinical vaccines,the establishment of diagnostic methods and the screening of porcine rotavirus vaccine candidate strains,and also enrich the epidemiological data of porcine rotavirus disease in Guizhou province.
