摘  要:为了解甘肃省地方猪场猪繁殖与呼吸综合征病毒(Porcine reproductive and respiratory syndrome virus, PRRSV)的感染情况,对2015-2020年采集的20个猪场332份疑似PRRSV感染猪的样品,首先进行血清学检测,然后通过RT-PCR方法开展病原检测,扩增PRRSV的ORF5基因;测序后利用DNA Star软件分析获得的ORF5基因及其编码的GP5氨基酸与国内外不同PRRSV毒株的遗传进化关系。血清学检测结果阳性率为93.07%,扩增获得的PRRSV ORF5基因,经核苷酸序列比对属于PRRSV美洲型,有NADC30-like毒株和NADC34 like毒株。结果表明,当前PRRSV不同流行毒株存在一定的遗传差异,为甘肃省PRRS防控提供了科学依据。In order to understand the infection status of porcine reproductive and respiratory syndrome virus(PRRSV)in local pig farms in Gansu province,332 samples of suspected PRRSV-infected pigs from 20 pig farms collected from 2015 to 2020 were firstly subjected to serological testing.The ORF5 gene was amplified by RT-PCR and sequenced,and the genetic evolution of the ORF5 gene and the GP5 amino acids encoded by it were analyzed by DNA Star software in relation to different PRRSV strains at home and abroad.The results showed that the positive serological test result was 93.07%,and the ORF5 gene of PRRSV obtained by amplification belonged to PRRSV American type by nucleotide sequence matching,with NADC30-like and NADC34-like strains.The results of the study showed that there are some genetic differences between the different prevalent strains of PRRSV,which provides some theoretical basis for the prevention and control of PRRS in Gansu province.
