摘 要:永生化细胞是研究细胞如何增殖、分化、凋亡过程及细胞衰老现象等机理的理想细胞模型。研究建立有效促进正常细胞永生化的生物学方法,相对于由某些基因突变、理化刺激及病毒、原癌基因直接诱导刺激等方法构建的永生化细胞系,应用端粒酶逆转录酶(telomerase reverse transcriptase, TERT)重新激活端粒酶活性来维持细胞增殖能力的方法被证明是安全有效的。论文阐述了近些年TERT所构建的永生化细胞系在组织工程、临床治疗、基础研究等方面的应用,总结了TERT构建的永生化细胞系对人和动物疾病相关研究起到的作用,并对其在细胞农业的应用价值进行了展望。Immortalized cell is an ideal cell model to study the mechanism of cell proliferation,differentiation,apoptosis and cell aging,which is conducive to basic research and bioengineering.Over the years,scholars have established a variety of biological methods to effectively promote the immortalization of normal cells.Compared with the immortalized cell lines constructed by some gene mutation,physical and chemical stimulation,and direct induction and stimulation of viruses and protooncogenes,the method of using telomerase reverse transcriptase(TERT)to reactivate telomerase activity to maintain cell proliferation has been proved to be safe and effective.In this paper,the applications of immortalized cell lines constructed by TERT in tissue engineering,clinical treatment and basic research in recent years are described,the role of immortalized cell lines constructed by TERT in the research of human and animal diseases is summarized,and its application value in cell agriculture is prospected.
