摘 要:目的 :探讨山茶籽对去卵巢大鼠成骨细胞雌激素受体α的表达和骨组织影响。方法 :12月龄SD雌性大鼠随机分成假手术组(只切除卵巢旁一块脂肪垫)、去卵巢模型组(模型组,腰背部切口摘除两侧卵巢)、山茶籽醇提物治疗组(治疗组,用山茶籽提取物灌胃)。取大鼠右侧股骨头1 cm用于脱钙后包埋切片观察骨组织形态结构,取大鼠左侧股骨头用于熏干扫描测定其骨密度。差速贴壁法分离采集成骨细胞,实时荧光定量PCR测定雌激素受体α mRNA的表达。结果 :光镜下见模型组股骨头骨小梁疏散、细小,周围皮质变薄,骨量减少;骨密度和第3腰椎骨骨细胞雌激素受体α mRNA的表达量均明显低于假手术组和治疗组,差异均有统计学意义。治疗组股骨头骨小梁变粗、变密,周围皮质变厚,骨量增加,骨密度和第3腰椎骨骨细胞雌激素受体α mRNA表达明显高于模型组,差异均有统计学意义。结论 :山茶籽醇提物能增加去卵巢大鼠骨密度和促进雌激素受体α mRNA的表达,骨组织病变改善良好。Objective:To investigate the effects of camellia seeds on the expression of estrogen receptorαand bone tissue in ovariectomized female rats.Methods:12-month-old Sprague-Dawley female rats were divided into three groups:a sham ovariectomy group,an ovariectomized group(OVX),and a camellia seed extract(OVX+Ts)group.One centimeter of the right femoral head was removed for decalcification and embedded section to observe the bone morphology and structure.The left femoral head of rats was taken for fumigation and scanning to determine the bone mineral density.The spinal bone was isolated for the isolation and collection of osteoblasts.Estrogen receptorαmRNA expression was detected by real-time PCR.Results:Microscopically,the trabecular bone of the femoral head in the OVX group was evacuated and small.The surrounding cortex was thinner,and bone mass decreased.The bone mineral density in the OVX group was significantly lower than that in the other two groups,and the expression of estrogen receptorαmRNA in osteocytes of the spinal bone in the OVX was also significantly lower than that in the sham group and OVX+Ts group.The trabecular bone of the OVX+Ts group became thicker and denser.The surrounding cortex became thicker,and the bone mass increased.The scanning bone mineral density of the OVX+Ts group was significantly higher than that of the OVX group,and the expression of estrogen receptorαmRNA in osteocytes of the backbone of the OVX+Ts group was also significantly higher than that in the sham group.Conclusion:The extract of camellia seed can increase bone mineral density,promote the expression of estrogen receptorαmRNA in OVX rats,and improves tissue structure of bone in OVX rats.
