摘  要:目的:构建具有缓释功能的β-环糊精包合弥可保丝素蛋白神经移植物,为临床修复周围神经缺损提供新的理论和实验依据。方法:(1)基于计算机模拟判断包合位点。(2)通过酰胺反应,构建环糊精丝素蛋白移植物。(3)通过缓释曲线研究弥可保的释放规律。(4)通过施万细胞的增殖实验和牛蛙坐骨神经缺损试验,验证移植物的生物相容性和对缺损神经的修复效果。结果:施万细胞的CCK-8实验表明移植物具有良好的体外生物相容性;移植物植入牛蛙缺损的坐骨神经,3 d后坐骨神经表现出一定的电生理功能恢复,说明移植物能促进牛蛙坐骨神经再生。结论:该神经移植物能有效缓释弥可保,具有良好的生物相容性,并能促进牛蛙缺损神经功能修复,为开发功能性人工神经支架提供重要策略。Objective:To construct mecobalamin/β-cyclodextrin complex loaded nerve graft with sustained release function,and provide a new theoretical and experimental basis for clinical repair of peripheral nerve defects.Methods:(1)The inclusion sites were determined by computer simulation.(2)Cyclodextrin-silk fibroin grafts were constructed through amide reaction.(3)The percentage of mecobalamin released from the grafts against time intervals was determined to study the release behavior.(4)Schwann cell proliferation test and bullfrog sciatic nerve defect test were used to verify the biocompatibility of the graft and the repair effect of the nerve defect.Results:CCK-8 assay of Schwann cells showed that the grafts had good biocompatibility in vitro.The graft was implanted into the damaged sciatic nerve,the sciatic nerve showed a certain recovery of electrophysiological function after 3 days,indicating that the graft could promote the regeneration of the sciatic nerve in bullfrogs.Conclusion:This nerve graft can effectively release mecobalamin,has good biocompatibility,and can promote the functionrepair of nerve defects in bullfrogs,which provides an important strategy for developing functional of artificialnervescaffolds.
