摘  要:应用旋毛虫混合排泄分泌蛋白(ES)和家兔IgG分别偶联铕(Eu)纳米颗粒(EuNPs)制备2个荧光探针,即EuNPs-ES探针和EuNPs-Rabbit IgG探针。将小鼠抗猪IgG与山羊抗家兔IgG分别包被于检测线与质控线上,制备时间分辨荧光免疫层析试纸条。采用15头不同剂量人工感染旋毛虫猪的全血评估研制的试纸条性能。结果显示,用试纸条检测感染200、400、1000、10000条旋毛虫肌幼虫的猪全血样品时,结果均为阳性。而使用集样消化法消化对应的猪膈肌样本时,低剂量(200条)未全部检出。以上结果表明,本研究中建立的时间分辨荧光免疫层析试纸条方法相比于集样消化法具有更高的灵敏度,且操作简便,仅需5~10 min即可出结果,本研究成功建立了一种用于现场快速检测旋毛虫病的方法。Two fluorescent probes were prepared by coupling europium(Eu)nanoparticles(EuNPs)with excretory-secretory proteins(ES)and rabbit IgG respectively,including EuNPs-ES probes and EuNPsrabbit IgG probes.Mouse anti-pig IgG and goat anti-rabbit IgG were coated on the test line and control line respectively.The time-resolved fluorescence immunochromatographic strip was successfully constructed.The whole blood of 15 pigs that were artificially infected with Trichinella spiralis at different doses was used to evaluate the performance of the developed strip.The results showed that the strips were 100%positive for whole blood from swine infected with 200,400,1000,and 10000 muscle larvae.The results of the digestion method for diaphragmatic muscle samples showed that the low dose(200 muscle larvae)was not all detected.The above results indicate that the time-resolved fluorescence immunochromatographic strip method has higher sensitivity than the digestion method,and it is simple to operate and only takes 5—10 min to get results.This study successfully established a method for the rapid detection of trichinellosis in the field.
