摘  要:以牛肾细胞(MDBK)细胞为研究对象,MDBK细胞接种牛冠状病毒(BCoV),应用Real-time PCR检测茯苓多糖对BcoV N基因表达水平及BcoV诱导的MDBK细胞中RLR干扰素信号通路上相关细胞因子表达水平的影响。结果,茯苓多糖可抑制BCoV N基因的表达;茯苓多糖可促进RLR通路细胞因子RIG-I、MDA5、MAVS、TBK1、IRF3及IFN-β的表达,并缓解BCoV对宿主细胞RIG-I、MDA5、MAVS、TBK1、IRF3及IFN-β细胞因子的前期抑制。结果表明,茯苓多糖可调节RLR通路细胞因子表达水平,抑制BCoV病毒的复制。这一研究结果为茯苓多糖治疗BCoV引起的犊牛腹泻提供了理论依据。To study the inhibitory effect of Poria cocos polysaccharide on bovine coronavirus(BCoV),MDBK cells were inoculated with BCoV,the real-time PCR was used to detect the expression level of Poria cocos polysaccharide on BcoV N gene and to analyze the expression level of related cytokines in RLR interferon signaling pathway in BCoV-induced MDBK cells.In result,Poria cocos polysaccharide could inhibit the expression of BCoV N gene.Poria cocos polysaccharide can promote the expression of RIG-I,MDA5,MAVS,TBK1,IRF3 and IFN-βin RLR pathway and alleviate the prophase inhibition of RIG-I,MDA5,MAVS,TBK1,IRF3 and IFN-βin host cells by BCoV.In conclusion,Poria cocos polysaccharide can inhibit the replication of BCoV virus by regulating the expression level of cytokines in RLR pathway,which provide a theoretical basis for the treatment of BCoV-induced calf diarrhea with Poria cocos polysaccharide.
