丝胶对地中海水牛精液冷冻保存效果的影响研究

摘  要:精液冷冻保存过程中精子易受氧化应激影响而导致解冻后精液品质下降。丝胶具有良好的抗氧化能力,在精液稀释液中添加丝胶可有效抑制活性氧积累导致的脂质过氧化,减缓精子氧化应激。为了解添加丝胶对地中海水牛精液冷冻保存效果的影响,本研究在冷冻稀释液中添加不同浓度(0、0.25%、0.5%、1%)的丝胶,进行精液冷冻保存。检测冻后精子活力、质膜完整性、顶体完整性及精浆中谷胱甘肽过氧化酶(GSH-Px)、总抗氧化能力(T-AOC)和丙二醛(MDA)水平。结果表明,与未处理组相比,添加0.5%丝胶可提高地中海水牛冻后精子活力、质膜完整性、顶体完整性、GSH-Px酶活性、T-AOC水平(P<0.05),MDA含量显著降低(P<0.05)。0.25%和1%丝胶添加组精子冻后活力、质量参数和抗氧化能力无显著性影响(P>0.05)。稀释液中添加0.5%丝胶时,冻后精子抗冻标志物HSP70蛋白表达水平极显著高于未添加组(P<0.01)。结果表明,在精液稀释液中加入0.5%丝胶可提高精液的抗氧化能力,使精子免受氧化应激损伤,有效改善地中海水牛精液冷冻后的品质。Sperm is susceptible to oxidative stress during semen cryopreservation,resulting in the decline of semen quality after thawing.Sericin has good antioxidant capacity,adding it to semen diluent can effectively inhibit lipid peroxidation caused by the accumulation of reactive oxygen species and slow down the oxidative stress of sperm.In order to understand the effect of sericin on buffalo semen cryopreservation,in this study,sericin with different concentrations(0,0.25%,0.5%,1%)was added to the semen freezing extender,the sperm motility,plasma membrane integrity and acrosome integrity,glutathione peroxidase(GSH-Px),total antioxidant capacity(T-AOC),malondialdehyde(MDA)oxidative stress indicators in seminal plasma of frozen-thawed semen were detected.The results showed that compared with the control group,the addition of 0.5%sericin significantly increased sperm motility,plasma membrane integrity and acrosome integrity,GSH-Px enzyme activity and T-AOC level(P<0.05),and significantly decreased the content of MDA in seminal plasma(P<0.05).There was no difference in the sperm vitality,quality and anti-inflammatory ability in the 0.25%and 1%sericin adding group(P>0.05).When 0.5%sericin was added to the freezing extender,the expression level of HSP70 protein,the antifreeze marker of frozen sperm,was significantly increased compared with the control group(P<0.01).In summary,the addition of 0.5%sericin to semen freezing extender can improve the antioxidant capacity of semen,protect sperm from oxidative stress damage,and effectively improve the quality of frozen semen of Mediterranean buffalo.
