摘 要:目的苗药“杆努尽烟”对慢性支气管炎(CB)大鼠肺组织激活蛋白-1(AP-1)因子以及炎症递质表达的影响。方法将50只SD大鼠,按照随机数表法分为空白对照组、模型组、阳性对照组、“杆努尽烟”高剂量组(8 g/kg)、“杆努尽烟”低剂量组(2 g/kg),除空白对照组外均采用香烟烟熏联合脂多糖气管内滴注法构建CB大鼠模型。造模成功后,各组连续给药28 d,分别予各大鼠等体积生理盐水、“杆努尽烟”高低剂量、桂龙咳喘宁等药物灌胃,观察各组大鼠炎症情况。28 d后,提取肺组织,观察肺部炎症病理改变,检测肺组织中AP-1(c-jun、c-fos)的蛋白含量、mRNA表达、肿瘤坏死因子-α(TNF-α)、白介素-8(IL-8)水平。结果与空白对照组比较,其余各组大鼠肺组织均受到损伤,有炎症细胞浸润,各组大鼠c-jun、c-fos蛋白表达以及mRNA水平、TNF-α、IL-8表达均有所增加(P<0.05);与模型组比较,各给药组肺组织损伤明显减轻,周围炎症细胞明显减少,且c-jun、c-fos蛋白表达以及mRNA水平、TNF-α、IL-8含量均有大幅度降低,差异有统计学意义(P<0.05),其中“杆努尽烟”高剂量组降低更为显著(P<0.05)。结论苗药“杆努尽烟”可能下调AP-1蛋白的表达,降低下游炎症因子IL-8、TNF-α的产生释放,从而改变肺部炎症病理状态,以减轻呼吸系统症状。Objective To investigate the effect of the Miao medicine Gannu Jinyan(cherbnex jenl vieeb in Hmong language)on the expression of activator protein-1(AP-1)factor and inflammatory mediators in the lung tissue of rats with chronic bronchitis(CB).Methods A total of 50 SD rats were randomly divided into a blank group,a model group,a positive control group,a high-dose Gannu Jinyan group(8 g/kg)and a low-dose Gannu Jinyan group(2 g/kg).The rat model of CB was constructed in these groups except the blank group by fumigation combined with intratracheal drip of lipopolysaccharide.After successful modeling,each group was gavaged with normal saline of equivalent volume,high and low doses of Gannu Jinyan and Guilong Kechuanning for 28 d,and the inflammatory situation in each group was observed.After 28 d,lung tissues were extracted for observation of the pathological changes of lung inflammation,and measurement of the protein content in AP-1(c-jun,c-fos),mRNA expression,and tumor necrosis factor-α(TNF-α)and interleukin-8(IL-8)levels.Results Except the blank group,the other groups had damaged lung tissues with inflammatory cell infiltration,and increased expression of c-jun and c-fos protein and levels of mRNA,TNF-αand IL-8(P<0.05).Compared with the model group,each administration group had significantly alleviated lung tissue damage,significantly reduced surrounding inflammatory cells,and statistically decreased expression of c-jun and c-fos proteins and levels of mRNA,TNF-αand IL-8,with statistically significant differences(P<0.05).Such reduction was more significant in the high-dose Gannu Jinyan group(P<0.05).Conclusion The Miao medicine Gannu Jinyan may down-regulate the expression of AP-1 protein and reduce the generation and release of downstream inflammatory factors IL-8 and TNF-α,thereby changing the pathological state of lung inflammation and alleviating respiratory symptoms.
