硼酸钠对小鼠的急性毒性实验研究

摘　　要：目的探讨硼酸钠单次经口给药对小鼠的急性毒性。方法根据预实验结果将C3H/HeN小鼠分为2.00、2.40、2.88、3.46、4.15、4.98和5.97 mg/g(Bliss法)7个硼酸钠实验组,对照组给予蒸馏水。灌胃均为最大给药容积,1 d内经口给药1次。观察14 d,记录小鼠的临床状态,死亡率和体重变化,并进行器官指标评价和病理检查。结果实验组小鼠表现出蜷卧少动、气短、毛发松散和粪便不成形,部分小鼠在1 h内死亡。解剖可见胃肠胀气,其他脏器未见异常。组织病理学结果显示脑、心、脾未见异常变化;肝细胞水肿、中央静脉扩张、瘀血;肺泡上皮结构正常,肺泡间隔毛细血管扩张外渗,淋巴细胞浸润,少量红细胞从肺泡腔漏出;肾小球及肾小管间质瘀血。硼酸钠雄性和雌性小鼠的LD50分别为3.794和3.646 mg/g。结论硼酸钠属于低毒化合物;急性硼酸钠中毒对小鼠肝脏、肺脏和肾脏有明显的损伤和毒性作用。Objective To investigate the acute toxicity of single oral administration of sodium borate to mice.Methods 7 experimental groups of C3H/HeN mice with sodium borate at 2.00,2.40,2.88,3.46,4.15,4.98 and 5.97 mg/g(Bliss method)respectively were set according to the pre-experimental results,and a control group was set and given distilled water.The maximum administration dose was used for gavage,and the drug was administered orally once per day.The clinical status,mortality and body weight changes of mice were recorded,and organ index assessment and pathological examination were performed during the 14 d of observation.Results The mice in the experimental groups presented curled up and inactivity,shortness of breath,loose hair and shapeless stool,and some of them died within 1 h.Gastrointestinal distention was seen through autopsy,and no abnormalities were observed in other organs.Histopathological findings showed no abnormal changes in brain,heart and spleen.Hepatocellular edema,central venous dilatation and stasis were observed.Normal alveolar epithelial structure,dilated and extravasated capillaries in the alveolar septum,lymphocytic infiltration,and a small amount of red blood cells leaking from the alveolar cavity were observed.And there was glomerular and tubular interstitial stasis.The median lethal dose(LD50)of sodium borate in male and female mice were 3.794 and 3.646 mg/g,respectively.Conclusion Sodium borate is a compound with low toxicity,while acute sodium borate poisoning has obvious damage and toxicity on liver,lung and kidney of mice.
