摘 要:目的探讨丹参酮Ⅱ_(A)对人肺腺癌细胞A549增殖、迁移、侵袭、凋亡的影响。方法实验分为空白对照组(A组,等体积培养基),顺铂组(B组,10μg/mL顺铂),丹参酮Ⅱ_(A)低、中、高剂量组(C1组、C2组、C3组,5,10,20μmol/L),各组细胞以加入相应药物或培养基处理。采用MTT法检测细胞增殖情况,划痕实验法观察细胞迁移情况,Transwell法检测细胞侵袭力,流式细胞术检测细胞凋亡情况,实时荧光定量聚合酶链反应(RT-qPCR)法及Western blot法检测细胞基质金属蛋白酶(MMP)-2、MMP-9、整合素(integrin)α2、integrinβ1、细胞周期蛋白B1(Cyclin B1)、半胱氨酸蛋白酶3(Caspase-3)mRNA和蛋白的表达水平。结果与A组比较,B组及C1组、C2组、C3组细胞处理24,48,72 h时的增殖率均显著降低,24 h时的迁移细胞数及侵袭细胞数均显著减少,凋亡率均显著升高(P<0.05)。与A组比较,B组及C1组、C2组、C3组细胞处理24 h的G0/G1期细胞占比均显著升高,S期、G2/M期细胞占比均显著降低(P<0.05)。与A组比较,B组及C1组、C2组、C3组细胞处理24 h的MMP-2,MMP-9,integrinα2,integrinβ1,Cyclin B1的mRNA和蛋白表达水平均显著降低,Caspase-3 mRNA和蛋白表达水平均显著升高(P<0.05)。结论丹参酮Ⅱ_(A)可抑制A549细胞的增殖、侵袭、迁移,并促进其凋亡,其作用机制可能与下调MMP-2,MMP-9,integrinα2,integrinβ1,Cyclin B1的表达和上调Caspase-3的表达有关。Objective To investigate the effects of tanshinoneⅡ_(A)on the proliferation,migration,invasion and apoptosis of human lung adenocarcinoma A549 cells.Methods The A549 cells were divided into the blank control group(group A,equal volume of culture medium),the cisplatin group(group B,10μg/mL cisplatin),the tanshinoneⅡ_(A)low-,medium-and high-dose groups(groups C1,C2 and C3,5,10,20μmol/L).Cells in each group were treated with the corresponding drug or culture medium.Cell proliferation was detected by the MTT method,cell migration was observed by the scratch assay,cell invasion was detected by the Transwell method,and apoptosis was detected by the flow cytometry.The expression levels of matrix metalloproteinase(MMP)-2,MMP-9,integrinα2,integrinβ1,Cyclin B1 and Caspase-3 mRNA and proteins were detected by the real-time fluorescence quantitative polymerase chain reaction(RT-qPCR)and Western blot.Results Compared with those in the group A,the proliferation rate of cells in the groups B,C1,C2 and C3 at 24,48,and 72 h of treatment decreased significantly,the numbers of migrating cells and invading cells in the groups B,C1,C2 and C3 at 24 h of treatment decreased significantly,and the apoptosis rate in the groups B,C1,C2 and C3 at 24 h of treatment increased significantly(P<0.05).Compared with those in the group A,the percentage of G0/G1-phase cells in the groups B,C1,C2 and C3 at 24 h of treatment increased significantly,while the percentage of S-phase and G2/M-phase cells in the groups B,C1,C2 and C3 at 24 h of treatment decreased significantly(P<0.05).Compared with those in the group A,the expression levels of MMP-2,MMP-9,integrinα2,integrinβ1 and Cyclin B1 mRNA and proteins in the groups B,C1,C2 and C3 at 24 h of treatment decreased significantly,while the expression levels of Caspase-3 mRNA and protein in the groups B,C1,C2 and C3 at 24 h of treatment increased significantly(P<0.05).Conclusion TanshinoneⅡ_(A)can inhibit the proliferation,invasion,migration of A549 cells,and can promote the apoptosis.Its mech
