摘  要:目的探讨山莴苣素对MDA-MB-453乳腺癌细胞增殖和迁移的作用。方法实验分为对照组和低、中、高剂量山莴苣素实验组。对照组细胞给予常规培养,低、中、高剂量山莴苣素实验组分别用10、20和30μmol·L^(-1)山莴苣素处理MDA-MB-453乳腺癌细胞24 h。以划痕愈合实验评估细胞的迁移能力;以蛋白质印迹法检测细胞中磷脂酰肌醇-3-激酶/蛋白激酶B(PI3K/Akt)信号通路相关蛋白表达水平。结果对照组和低、中、高剂量山莴苣素组的迁移率分别为(29.35±2.99)%、(21.44±1.14)%、(18.83±0.49)%和(10.73±0.43)%;Akt蛋白的相对表达水平分别为1.00±0.04、0.94±0.05、0.92±0.05和1.09±0.03;p-Akt蛋白的相对表达水平分别为1.00±0.01、0.43±0.04、0.30±0.03和0.31±0.03;PI3K蛋白相对表达水平分别为1.00±0.04、1.03±0.19、1.14±0.12和1.09±0.08;p-PI3K蛋白相对表达水平分别为1.00±0.02、1.06±0.15、1.12±0.08和1.09±0.07。以上指标,低、中、高剂量山莴苣素实验组与对照组比较,差异均有统计学意义(均P<0.05)。结论山莴苣素通过抑制PI3K/Akt信号通路,抑制MDA-MB-453乳腺癌细胞增殖和迁移。Objective To investigate the effect of lactucin on the proliferation and migration of MDA-MB-453 breast cancer cells.Methods The experiment was divided into control group and low,medium and high dose groups.MDA-MB-453 breast cancer cells were treated with 10,20 and 30μmol·L^(-1 )lactucin in the low,middle and high dose groups for 24 h,respectively.The migration ability of cells was evaluated by scratch healing assay.The expression levels of phosphatidylinositol-3-kinase/protein kinase B(PI3K/Akt)signaling pathway-related proteins in cells were detected by Western blotting.Results The migration rates of the control group and the low,middle and high doses groups ware(29.35±2.99)%,(21.44±1.14)%,(18.83±0.49)%and(10.73±0.43)%,respectively;the relative expression levels of Akt protein were 1.00±0.04,0.94±0.05,0.92±0.05 and 1.09±0.03,respectively;the relative expression levels of p-Akt protein were 1.00±0.01,0.43±0.04,0.30±0.03 and 0.31±0.03,respectively;the relative expression levels of PI3K protein were 1.00±0.04,1.03±0.19,1.14±0.12 and 1.09±0.08,respectively;the relative expression levels of p-PI3K protein were 1.00±0.02,1.06±0.15,1.12±0.08 and 1.09±0.07,respectively.Compared with the control group,the above indexes in the low,middle and high dose groups were significantly different(all P<0.05).Conclusion Lactucin inhibits proliferation and migration of MDA-MB-453 breast cancer cells by inhibiting PI3K/Akt signaling pathway.
