益气明目颗粒含药血清对氯化钴诱导缺氧损伤RGC-5细胞的保护作用

摘　　要：目的观察益气明目颗粒含药血清对氯化钴(CoCl2)诱导缺氧损伤小鼠视网膜神经节细胞(RGC-5)的保护作用。方法采用CoCl2构建RGC-5缺氧损伤模型,分别用CCK-8法检测细胞活力,流式细胞术检测细胞凋亡水平,生物透射电镜观察细胞超微结构改变,试剂盒检测细胞内谷胱甘肽过氧化物酶(GSH-Px)和超氧化物歧化酶(SOD)含量的变化,RT-PCR检测缺氧诱导因子-1α(HIF-1α)、半胱天冬酶-3(Caspase-3)、B淋巴细胞瘤因子相关x蛋白(Bax)、B细胞淋巴瘤基因-2(Bcl-2)相关mRNA表达水平。结果不同浓度含药血清组细胞存活率均高于缺氧模型组(P<0.05,P<0.01),且2.5%、5%含药血清组细胞存活率显著高于同浓度空白血清组(P<0.05),抑制细胞凋亡,减轻细胞超微结构损伤,提高细胞GSH-Px和SOD水平(P<0.01);与模型组相比,2.5%、5%浓度含药血清能够下调细胞HIF-1α、Caspase-3、Bax的表达(P<0.05,P<0.01),升高Bcl-2的表达(P<0.01)。结论益气明目颗粒含药血清能够通过抗氧化应激作用减轻CoCl2诱导的RGC-5缺氧损伤。Objective To observe the protective effect of Yiqi Mingmu granule containing serum on retinal ganglion cells(RGC-5)induced by cobalt chloride(CoCl2).Methods The hypoxic injury model of RGC-5 was established by CoCl2.The cell via-bility was detected by CCK-8 method,the level of apoptosis was detected by flow cytometry,the ultrastructural changes of cells were observed by biological transmission electron microscope,and the contents of glutathione peroxidase(GSH-Px)and super-oxide dismutase(SOD)were detected by kit.The expression levels of hypoxia inducible factor-1α(HIF-1α),caspase-3(Caspase-3),B-lymphocyte tumor factor-associated x protein(Bax)and B-cell lymphoma gene-2(Bcl-2)-related mRNA were detected by RT-PCR.Results The cell survival rate in different concentrations of drug-containing serum group was higher than that in hypoxia model group(P<0.05),and the cell survival rate in 2.5%and 5%drug-containing serum group was significantly higher than that in the same concentration blank serum group(P<0.05).It can inhibit cell apoptosis,reduce cell ultrastructural damage and increase the levels of GSH-Px and SOD.Compared with the model group,2.5%and 5%concentration of drug-containing serum could down-regulate the expression of HIF-1α,Caspase-3 and Bax and increase the expression of Bcl-2(P<0.05).Conclusion The drug-containing serum of Yiqi Mingmu granule can alleviate the anoxic inju-ry of RGC-5 induced by CoCl2 through antioxidant stress.
