摘  要:目的观察黄龙舒喘汤干预哮喘模型大鼠气道高反应的作用,探讨干预作用与S1P/S1PR通路的相关性。方法将60只Wistar大鼠随机分为6组,即对照组、模型组、激素组、黄龙舒喘汤低、中、高剂量组。根据分组情况,给予相应的干预措施,并采用相应的方法检测。结果经黄龙舒喘汤干预后,哮喘大鼠症状有不同程度减轻,气道阻力值(RI)均较模型组降低(P<0.05),能显著降低肺泡灌洗液(BALF)中总细胞、嗜酸性粒细胞(EOs)和中性粒细胞(NEUT)计数(P<0.05);肺组织病理可见激素组和各HLSC组可见不同程度的气管舒张、分泌物减少、气管周围炎症细胞减少;SphK2表达显著降低(P<0.05),S1PR2、S1PR3表达较模型组显著下调(P<0.05)。结论采用黄龙舒喘汤干预哮喘大鼠模型,能减轻气道阻力,改善气道高反应性(AHR)和气道炎症细胞浸润,降低肺组织中表达增多的SphK2,其机制可能与抑制S1PR2/S1PR3介导的通路有关。Objective To observe the effect of Huanglong Shuchuan Decoction on airway hyper reactivity in asthmatic rats,and explore the correlation between the intervention and S1P/S1PR pathway.Methods Sixty Wistar rats were randomly divided into 6 groups,namely,blank group,model group,hormone group and low,middle and high dose groups of Huanglong Shuchuan Decoc-tion.According to the grouping situation,the corresponding intervention measures were given,and the corresponding methods were used for detection.Results After the intervention of Huanglong Shuchuan Decoction,the symptoms of asthma rats were re-lieved in different degrees,and the airway resistance(RI)was lower than that of the model group(P<0.05),which could sig-nificantly reduce the total cell,eosinophil(EOs)and neutrophil(NEUT)counts in bronchoalveolar lavage fluid(BALF)(P<0.05).Lung histopathology showed that different degrees of tracheal relaxation,secretion reduction and inflammatory cells around trachea were observed in hormone group and HLSC groups.The expression of SphK2 decreased significantly(P<0.05),and the expression of S1PR2 and S1PR3 decreased significantly compared with that of the model group(P<0.05).Conclusion The in-tervention of Huanglong Shuchuan Decoction in asthma rat model can reduce airway resistance,improve airway hyper reactivity(AHR)and infiltration of airway inflammatory cells,and reduce the increased expression of SphK2 in lung tissue.The mecha-nism may be related to the inhibition of S1PR2/S1PR3-mediated pathway.
