摘 要:为开展源自皂荚的3种中药(大皂角、猪牙皂和皂角刺)质量控制研究,该文利用超高效液相色谱-三重四极杆-线性离子阱质谱(UHPLC-Q-Trap-MS),构建了多重反应监测(MRM)方法,采用ACQUITY UPLC BEH C18色谱柱(2.1 mm×100 mm, 1.7μm),在40℃柱温下以0.1%甲酸水-乙腈为流动相、0.3 mL·min^(-1)的流速进行梯度洗脱,在31 min内同时实现了大皂角、猪牙皂和皂角刺中10种化学成分(saikachinoside A、locustoside A、荭草苷、花旗松素、牡荆素、异槲皮苷、木犀草苷、槲皮苷、槲皮素、芹菜素)的分离与含量测定。该方法可以快速、高效地测定大皂角、猪牙皂和皂角刺中以上10种化学成分的含量,各成分均呈现良好的线性关系(r>0.995),平均加样回收率为94.09%~110.9%。结果表明,大皂角中2种生物碱类成分质量分数(2.03~834.75μg·g^(-1))明显高于猪牙皂(0.03~10.41μg·g^(-1))和皂角刺(0.04~13.66μg·g^(-1));皂角刺中8种黄酮类成分的质量分数(0.54~2.38 mg·g^(-1))明显高于大皂角(0.08~0.29 mg·g^(-1))和猪牙皂(0.15~0.32 mg·g^(-1))。这些结果可为源自皂荚的中药质量控制提供参考。In order to conduct the quality control of three traditional Chinese medicines derived from Gleditsia sinensis(Gleditsiae Sinensis Fructus,GSF;Gleditsiae Fructus Abnormalis,GFA;Gleditsiae Spina,GS),a multiple reaction monitoring(MRM)approach was established based on ultra-high performance liquid chromatography-triple quadrupole-linear ion-trap mass spectrometry(UHPLC-Q-Trap-MS).Using an ACQUITY UPLC®BEH C18 column(2.1 mm×100 mm,1.7μm),gradient elution at 40℃with 0.1%formic acid in water-acetonitrile as the mobile phase running at 0.3 mL·min^(-1),the separation and precise quantification of 10 chemical constituents(e.g.,saikachinoside A,locustoside A,orientin,taxifolin,vitexin,isoquercitrin,luteolin,quercitrin,quercetin,and apigenin)in GSF,GFA,and GS,was enabled within 31 min.The established method could quickly and efficiently determine the contents of 10 chemical components in GSF,GFA,and GS.All components showed good linearity(r>0.995),and the average recovery was 94.09%-110.9%.The results showed that,the contents of two alkaloids in GSF(2.03-834.75μg·g^(-1))were higher than those in GFA(0.03-10.41μg·g^(-1))and GS(0.04-13.66μg·g^(-1)),while the contents of eight flavonoid components in GS(0.54-2.38 mg·g^(-1))were higher than those of GSF(0.08-0.29 mg·g^(-1))and GFA(0.15-0.32 mg·g^(-1)).These results can provide reference to the quality control of G.sinensis-derived TCMs.
