摘  要:试验旨在研究日粮不同能量水平对乌金猪肌肉组织蛋白质翻译起始因子基因表达的影响。选取健康、体重约15 kg的乌金猪54头,随机分为3组,每组3个重复,每个重复6头猪。各组猪饲喂不同能量水平的日粮(11.84、12.97、14.18 MJ/kg),试验分15~30、31~60、61~100 kg 3个阶段。各阶段换料过渡期为7 d,分别于猪30、60、100 kg时分批屠宰。结果显示,高能量水平日粮可降低猪的瘦肉率和肌肉粗蛋白含量,增加肌内脂肪含量,降低蛋白质翻译起始因子基因的表达。屠宰体重为30 kg时,低能量组猪的真核起始因子-2B(eIF-2B)、真核起始因子-4E(eIF-4E)表达水平显著高于高能量组(P<0.05),真核起始因子-4B (eIF-4B)基因表达量显著高于中能量组(P<0.05)。屠宰体重为60 kg时,高能量组和中能量组猪的eIF-2B、eIF-4B、eIF-4E表达水平显著低于低能量组(P<0.05)。屠宰体重为100 kg时,高能量组猪的真核起始因子-4A (eIF-4A)的相对表达水平显著低于低能量组和中能量组(P<0.05),低能量组eIF-4E相对表达量显著高于中能量组和高能量组(P<0.05);中能量组的eIF-4E相对表达量显著高于高能量组(P<0.05)。研究表明,乌金猪背最长肌蛋白质沉积随日粮能量水平降低而升高,肌内脂肪含量反之,日粮不同能量水平可通过调节蛋白质合成翻译起始因子基因的表达,明显影响乌金猪肌肉组织蛋白质的合成。The experiment was to investigate the effect of different dietary energy levels on protein translation initiation factor gene expression in muscle tissue of Wujin pigs.Fifty-four healthy Wujin pigs with an average body weight of 15 kg were randomly divided into three groups with three replicates in each group and six pigs in each replicate.Pigs in each group were fed diets with different energy levels(11.84,12.97 and 14.18 MJ/kg),and the experiment was divided into three stages:15~30,31~60 and 61~100 kg.The refueling transition period of each stage was 7 d,and the animals were slaughtered in batches at 30,60,100 kg,respectively.The results showed that high energy level diet could reduce lean meat percentage and muscle crude protein content,increase intramuscular fat content of pigs,decrease protein translation initiation factor gene expression.At slaughter body weight of 30 kg,the expression levels of eIF-2B and eIF-4E in low energy group were significantly higher than those in high energy group(P<0.05),and the expression level of eIF-4B gene was significantly higher than that in medium energy group(P<0.05).When slaughter weight was 60 kg,the expression levels of e IF-2B,e IF-4B and e IF-4E in high and medium energy groups were significantly lower than those in low energy group(P<0.05).At slaughter weight of 100 kg,the relative expression level of e IF-4A in high energy group was significantly lower than that in low energy group and medium energy group(P<0.05),and the relative expression level of e IF-4E in low energy group was significantly higher than that in medium energy group and high energy group(P<0.05).The relative expression level of e IF-4E in medium energy group was significantly higher than that in high energy group(P<0.05).The study indicates that protein deposition in longissimus dorsi muscle of Wujin pigs increases with the decrease of dietary energy level,while intramuscular fat content is opposite.Different dietary energy levels can significantly affect protein synthesis in muscle tissue of Wujin
